【摘要】：目的:探討維生素E對(duì)錳中毒小鼠學(xué)習(xí)記憶及海馬區(qū)caspase-3表達(dá)的影響。方法:將60只雄性昆明小鼠隨機(jī)分為四組:對(duì)照組(CG)、維生素E組(VG)、錳中毒組(MG)、錳中毒加維生素E組(MVG),每組15只。采用腹腔注射氯化錳的方式制作錳中毒模型,Morris水迷宮觀察小鼠的學(xué)習(xí)記憶能力,免疫組織化學(xué)方法檢測(cè)caspase-3蛋白的表達(dá)。結(jié)果:Morris水迷宮定位航行實(shí)驗(yàn)顯示,CG、VG、MVG三組的逃避潛伏期比MG組明顯縮短,空間探索實(shí)驗(yàn)中,CG、VG、MVG三組穿越平臺(tái)的次數(shù)明顯多于MG組。免疫組織化學(xué)實(shí)驗(yàn)中,MG組海馬CA1區(qū)和DG區(qū)caspase-3的表達(dá)明顯多于CG組,MVG組海馬區(qū)caspase-3的陽(yáng)性細(xì)胞數(shù)明顯少于MG組。結(jié)論:維生素E干預(yù)可減少海馬區(qū)神經(jīng)細(xì)胞的凋亡,改善錳中毒小鼠的學(xué)習(xí)記憶能力。
[Abstract]:Objective: to investigate the effects of vitamin E on learning and memory and caspase-3 expression in hippocampus of manganese intoxicated mice. Methods: sixty male Kunming mice were randomly divided into four groups: control group, (CG), vitamin E group, (VG), manganese poisoning group, (MG), manganese poisoning group and vitamin E group (MVG), 15 rats in each group. The model of manganese poisoning was made by intraperitoneal injection of manganese chloride. Morris water maze was used to observe the learning and memory ability of mice. The expression of caspase-3 protein was detected by immunohistochemical method. Results the escape latency of the three groups was significantly shorter than that of the MG group, and the frequency of crossing the platform in the three groups was significantly higher than that in the MG group. The expression of caspase-3 in CA1 and DG was significantly higher in MVG group than in CG group. The number of caspase-3 positive cells in MVG group was significantly lower than that in MG group. Conclusion: vitamin E intervention can reduce apoptosis of hippocampal neurons and improve learning and memory ability of mice exposed to manganese.
