注射用氯唑西林鈉中氯唑西林噻唑酸的分離及含量檢測(cè)

【摘要】:目的:實(shí)現(xiàn)注射用氯唑西林鈉中氯唑西林噻唑酸的分離及含量檢測(cè)。方法:采用分子印跡-溶膠凝膠技術(shù),以氯唑西林噻唑酸為模板,在活化硅膠表面制備具有特異性的分子印跡聚合物,并對(duì)合成條件進(jìn)行優(yōu)化。采用掃描電鏡對(duì)聚合物進(jìn)行形貌表征。將聚合物作為固相萃取填料,結(jié)合高效液相色譜法對(duì)氯唑西林鈉中氯唑西林噻唑酸進(jìn)行分離及含量檢測(cè)。結(jié)果:當(dāng)模板分子、功能單體和交聯(lián)劑的摩爾比為1∶4∶28,溶劑為3 mL,催化劑為300μL,反應(yīng)溫度為30℃時(shí),聚合物的吸附量和印跡因子最佳。掃描電鏡結(jié)果顯示聚合物已成功接枝到硅膠表面。氯唑西林噻唑酸分子印跡聚合物和固相萃取相結(jié)合的方法,經(jīng)方法學(xué)驗(yàn)證,在0.5~50 mg·g~(-1)范圍內(nèi)呈現(xiàn)良好的線性關(guān)系(r=0.996 2),檢測(cè)限為0.07 mg·g~(-1)。結(jié)論:該方法的專屬性、準(zhǔn)確度和精密度均良好,能夠?qū)崿F(xiàn)氯唑西林鈉中微量雜質(zhì)氯唑西林噻唑酸的分離及含量檢測(cè)。
[Abstract]:Objective: to establish the separation and determination of chlorzocillin thiazole acid in chlorzocillin sodium for injection. Methods: molecularly imprinted polymers were prepared on activated silica gel surface by molecular imprinting and sol-gel technique using chlorzocillin thiazole acid as template and the conditions of synthesis were optimized. The morphology of the polymer was characterized by SEM. The polymer was used as solid phase extraction filler and the content of chlorzocillin in chlorzocillin sodium was determined by high performance liquid chromatography (HPLC).
