【摘要】：目的探討精脒對(duì)人結(jié)腸癌SW620細(xì)胞增殖及糖酵解的影響。方法以SW620細(xì)胞為研究對(duì)象,用0.625~2.5μmo L/L精脒(SPD)作用于細(xì)胞,細(xì)胞密度以104個(gè)/m L接種于96孔培養(yǎng)板,分別培養(yǎng)24 h后,MTT法檢測(cè)細(xì)胞增殖情況;同時(shí),取6孔板培養(yǎng)24 h的細(xì)胞上清液,用試劑盒檢測(cè)SW620細(xì)胞葡萄糖消耗及乳酸水平;細(xì)胞密度以1.2×10~5個(gè)/m L接種于6孔培養(yǎng)板中,培養(yǎng)24 h后加入不同濃度SPD,繼續(xù)培養(yǎng)24 h后,Western blot檢測(cè)缺氧誘導(dǎo)因子(HIF-1)、乳酸脫氫酶(LDHA)及葡萄糖轉(zhuǎn)運(yùn)蛋白1(GLUT1)表達(dá)的變化。結(jié)果與對(duì)照組比較,精脒各濃度組均能促進(jìn)SW620細(xì)胞的增殖(P0.01),且促進(jìn)作用呈劑量依賴性;精脒各濃度組作用后SW620細(xì)胞內(nèi)葡萄糖的消耗及乳酸含量明顯增多(P0.01),呈劑量依賴性;與對(duì)照組比較,LDHA、HIF-1及GLUT1蛋白表達(dá)水平隨著精脒濃度的升高而增加(P0.01),且呈劑量依賴性。結(jié)論精脒具有促進(jìn)人結(jié)腸癌SW620細(xì)胞增殖及糖酵解的作用。
[Abstract]:Objective to investigate the effect of spermidine on proliferation and glycolysis of human colon cancer SW620 cells. Methods SW620 cells were treated with 0.625 渭 mo L / L spermidine (SPD), and the cell density was 104 / mL inoculated on 96 well culture plate. After 24 h culture, cell proliferation was detected by MTT assay. The supernatant of 6-well plate cultured for 24 h was used to detect glucose consumption and lactic acid level of SW620 cells, the cell density was 1.2 脳 10 ~ 5 / mL and inoculated in 6-well culture plate. After 24 hours of culture, the expression of hypoxia inducible factor (HIF-1), lactate dehydrogenase (LDHA) and glucose transporter 1 (GLUT1) were detected by Western blot. Results compared with the control group, spermidine could promote the proliferation of SW620 cells (P0.01) in a dose-dependent manner, and the glucose consumption and lactic acid content in SW620 cells were increased in dose-dependent manner after the treatment of spermidine (P0.01). Compared with the control group, the expression of HIF-1 and GLUT1 protein increased with the increase of spermidine concentration (P0.01), in a dose-dependent manner. Conclusion spermidine can promote the proliferation and glycolysis of human colon cancer SW620 cells.
