耐酸性布拉氏酵母的篩選及高密度培養(yǎng)條件優(yōu)化

【摘要】:布拉氏酵母菌(Saccharomyces boulardii)是臨床上作為益生菌藥物治療腸道疾病使用的唯一一株酵母菌,作為微生態(tài)制劑,要保證其生物效果,必須能夠在胃腸道中保持一定的活菌數(shù),研究發(fā)現(xiàn)影響布拉氏酵母菌在胃腸道中存活的最主要因素是低p H,為提高布拉氏酵母菌在低p H條件下的存活率,該研究采用常壓室溫等離子體(ARTP)誘變技術對S.boulardii進行誘變,最終篩選出3株對低p H值耐受性較好的突變株。對突變株耐低p H穩(wěn)定性的研究結果表明,在傳代20次后,突變株YB-3具有較好的遺傳穩(wěn)定性,其存活率為51.79%。在5 L發(fā)酵罐中,對突變株YB-3高密度培養(yǎng)條件進行優(yōu)化,最終所得布拉氏酵母菌體干質量為58.79 g/L,比原始菌株提高了55.52%。
[Abstract]:(Saccharomyces boulardii) is the only yeast used clinically as a probiotic drug for the treatment of intestinal diseases. As a microecological preparation, to ensure its biological effect, it must be able to maintain a certain number of live bacteria in the gastrointestinal tract. It was found that the most important factor affecting the survival of Saccharomyces brasilii in the gastrointestinal tract was low pH. In order to improve the survival rate of Saccharomyces brasiliensis at low pH, the S.boulardii was mutagenic by plasma (ARTP) at room temperature at atmospheric pressure. Finally, 3 mutants with good tolerance to low pH were selected. The results showed that the mutant YB-3 had good genetic stability after 20 passages, and its survival rate was 51.79%. In a 5L fermenter, the high density culture conditions of mutant YB-3 were optimized, and the dry mass of yeast was 58.79 g / L, which was 55.52% higher than that of the original strain.
