HPLC法測(cè)定苯磺酸左旋氨氯地平片有關(guān)物質(zhì)及主要雜質(zhì)鑒別

【摘要】:目的:建立苯磺酸左旋氨氯地平片有關(guān)物質(zhì)測(cè)定方法,對(duì)主要雜質(zhì)進(jìn)行鑒別并采用雜質(zhì)對(duì)照品測(cè)定校正因子。方法:采用HPLC方法分離分析苯磺酸左旋氨氯地平片有關(guān)物質(zhì),使用Agilent XDB C18(250 mm×4.6 mm,5μm)色譜柱,流動(dòng)相為甲醇-乙腈-0.7%三乙胺溶液(取三乙胺7.0 m L,加水至1 000 m L,用磷酸調(diào)節(jié)p H至3.0±0.1)(35∶15∶50),流速1.0 m L·min-1,檢測(cè)波長(zhǎng)238 nm,柱溫30℃;采用2.3 g·L-1醋酸銨-甲醇(40∶60)為流動(dòng)相,利用LC-MS/MS方法進(jìn)行鑒別。結(jié)果:苯磺酸左旋氨氯地平與有關(guān)物質(zhì)能有效分離;LC-MS/MS鑒定了5種主要雜質(zhì);雜質(zhì)D(3-乙基-5-甲基-2-[(2-氨基乙氧基)甲基]-4-(2-氯苯基)-6-甲基吡啶-3,5-二羧酸)、雜質(zhì)E(二乙基-2-[(2-氨基乙氧基)甲基]-4-(2-氯苯基)-6-甲基-1,4-二氫吡啶-3,5-二羧酸)、雜質(zhì)F(二甲基-2-[(2-氨基乙氧基)甲基]-4-(2-氯苯基)-6-甲基-1,4-二氫吡啶-3,5-二羧酸)、氨氯地平乳糖加合物的定量限分別為9.3、10.4、5.1、19.1 ng,且在各自的線性范圍內(nèi)線性關(guān)系良好(r=1.000,n=5);平均回收率(n=9)分別為113.8%、101.6%、103.4%、107.9%;校正因子分別為2.0、1.1、1.0、3.0。結(jié)論:建立的方法可對(duì)苯磺酸左旋氨氯地平片有關(guān)物質(zhì)進(jìn)行定量測(cè)定。
