【摘要】：目的探索試驗樣品組氨酸含量對Ames試驗中測試菌株TA97 a、TA98、TA100、TA102和TA1535自發(fā)回變菌落數(shù)的影響。方法使用0.8、0.4、0.2、0.1和0.05 mg/ml 5個濃度的組氨酸水溶液作為受試物按平板摻入法進行Ames試驗。結(jié)果在一定濃度范圍內(nèi),隨著組氨酸劑量的增高,5個菌株的回變菌落數(shù)均逐漸增多。TA98和TA1535的回變菌落數(shù)分別在0.8和0.2 mg/ml時超過陰性對照組的2倍。所有測試菌株在0.2 mg/ml時均出現(xiàn)了與對照組間的差異有統(tǒng)計學(xué)意義。結(jié)論在進行Ames試驗前,對于受試物中游離組氨酸含量的判定十分必要;當(dāng)受試物中組氨酸含量高于0.2 mg/ml時,有可能造成"假陽性"結(jié)果的產(chǎn)生,建議采用以基因突變?yōu)闄z測終點的其他試驗進行替代。
[Abstract]:Objective to explore the effect of the content of histidine on the number of spontaneous retrogression colonies of TA97 TA98 TA100 TA102 and TA1535 in Ames test. Methods five concentrations of histidine (0. 8, 0. 4, 0. 2, 0. 1 and 0. 05 mg/ml) were used as samples for Ames test. Results in a certain concentration range, with the increase of the dose of histidine, the number of retrograde colonies of 5 strains increased gradually. The number of retrograde colonies of TA98 and TA1535 was 2 times higher than that of negative control at 0.8 and 0.2 mg/ml, respectively. All the tested strains were significantly different from the control at 0. 2 mg/ml. Conclusion it is necessary to determine the content of free histidine before Ames test, and when the content of histidine is higher than 0.2 mg/ml, it is possible to produce false positive results. It is recommended that other tests with gene mutation as the detection endpoint be used instead.
