用HPLC法測(cè)定人血漿中丙戊酸鈉的游離濃度

【摘要】:目的建立一種測(cè)定人血漿中丙戊酸鈉游離濃度的中空纖維離心超濾-高效液相色譜(HPLC)方法。方法血漿樣本經(jīng)中空纖維離心超濾技術(shù)預(yù)處理,經(jīng)2-溴甲基-2-萘乙酮衍生后進(jìn)行HPLC分析。色譜柱:Diamonsil RPC18(4.6 mm×150 mm,5μm),流動(dòng)相:甲醇∶水=80∶20;流速:1.0 m L·min~(-1);柱溫:30℃;檢測(cè)波長(zhǎng):251 nm?疾煸摲椒ǖ膶傩、標(biāo)準(zhǔn)曲線和定量下限、精密度與回收率、穩(wěn)定性。結(jié)果人血漿中游離的丙戊酸鈉在該條件下分離良好,在0.05~50.00μg·m L~(-1)內(nèi)線性關(guān)系良好(r=0.999 0),定量下限為0.01μg·m L~(-1),方法回收率為96.51%~101.22%,絕對(duì)回收率達(dá)到94.28%~99.15%,批內(nèi)、批間RSD均4.00%,在室溫放置10 h、凍融循環(huán)3次和冰凍21 d的穩(wěn)定性良好。結(jié)論本方法簡(jiǎn)便、準(zhǔn)確、靈敏,適用于人血漿中丙戊酸鈉的游離濃度分析。
