黃芩素-煙酰胺共晶結(jié)晶過程的熱力學(xué)研究

【摘要】:以黃芩素-煙酰胺共晶為模型藥物,研究共晶形成的熱力學(xué)。通過測定不同溫度下黃芩素在不同濃度煙酰胺溶液中的溶解度,探討黃芩素-煙酰胺共晶的絡(luò)合形式,計算相關(guān)熱力學(xué)參數(shù),并繪制不同溫度下的黃芩素-煙酰胺-乙酸乙酯三元相圖。結(jié)果表明,黃芩素-煙酰胺共晶在乙酸乙酯中符合物質(zhì)的量比1∶1溶液絡(luò)合模型,共晶形成反應(yīng)為熱力學(xué)自發(fā)反應(yīng)(ΔG°0),隨著溫度升高,Ksp逐漸增大,K11逐漸減小,反應(yīng)自發(fā)程度減弱,三元相圖中共晶區(qū)域減小,當(dāng)溫度達(dá)到T*=315 K后,ΔG°=0(ΔH°=-6.314×10-2k J/mol,ΔS°=-0.200 5 J/mol·K),自發(fā)反應(yīng)不再進(jìn)行,證明低溫有利于共晶的形成。根據(jù)三元相圖的非對稱特點(diǎn),設(shè)計了3種不以物質(zhì)的量比1∶1為出發(fā)點(diǎn)制備共晶的方法,DSC結(jié)果表明,3種方法制備得到的黃芩素-煙酰胺共晶均為熔點(diǎn)單一的純共晶。
[Abstract]:The thermodynamics of eutectic formation was studied using baicalin-nicotinamide eutectic as model drug. By measuring the solubility of baicalin in nicotinamide solution with different concentrations at different temperatures, the complexation of baicalein-nicotinamide eutectic was discussed, and the thermodynamic parameters were calculated. The ternary phase diagrams of baicalein-nicotinamide-ethyl acetate were drawn at different temperatures. The results showed that baicalein-nicotinamide eutectic was in accordance with the 1:1 solution complexation model in ethyl acetate. The eutectic reaction was thermodynamic spontaneous reaction (螖 G 擄0), and gradually decreased with the increase of temperature. The spontaneous reaction decreased and the eutectic region of ternary phase diagram decreased. When the temperature reached 315K, 螖 G 擄O (螖 H 擄-6.314 脳 10-2k J / mol, 螖 S 擄-0.200 5 J/mol K),) no longer occurred. It is shown that low temperature is favorable to eutectic formation. According to the asymmetric characteristics of ternary phase diagram, three kinds of eutectic methods were designed to prepare eutectic without the 1:1 mass ratio of substance. The DSC results showed that baicalein-nicotinamide eutectic was pure eutectic with a single melting point.
