【摘要】：目的:研究抗壞血酸對(duì)成骨細(xì)胞膜片BMP-2 mRNA表達(dá)的影響。方法:原代培養(yǎng)并鑒定大鼠成骨細(xì)胞,采用物理刮取法,體外構(gòu)建成骨細(xì)胞膜片;成骨細(xì)胞接種后,分別加入15、50、85 mg/L抗壞血酸,連續(xù)培養(yǎng)1~2周,RT-q PCR檢測(cè)成骨細(xì)胞膜片BMP-2 mRNA的表達(dá)。結(jié)果:獲得的細(xì)胞符合成骨細(xì)胞形態(tài)學(xué)特征特性�？蓸�(gòu)建管狀形態(tài)的成骨細(xì)胞膜片。組間比較,第1周和第2周成骨細(xì)胞膜片中BMP-2 mRNA表達(dá)量實(shí)驗(yàn)組均高于對(duì)照組,以50 mg/L組的表達(dá)量最高(第1周P0.05;第2周P0.05);組內(nèi)比較,各組表達(dá)量第2周均高于第1周(P0.05)。結(jié)論:抗壞血酸可促進(jìn)成骨細(xì)胞膜片BMP-2mRNA表達(dá)。
[Abstract]:Objective: to study the effect of ascorbic acid on the expression of BMP-2 mRNA in osteoblasts. Methods: primary culture and identification of rat osteoblasts were used to construct osteoblast membrane by physical curettage in vitro. After inoculation of osteoblasts, ascorbic acid (ascorbic acid) was added to the osteoblasts for 1 week and 2 weeks, respectively. The expression of BMP-2 mRNA in osteoblasts was detected by RT-Q PCR. Results: the obtained cells accord with the morphological characteristics of osteoblasts. The tubular osteoblast membrane can be constructed. The expression of BMP-2 mRNA in the first and second week of osteoblast in the experimental group was higher than that in the control group, the expression of BMP-2 mRNA in the 50 mg/L group was the highest (P0.05 in the first week; P0.05 in the second week); in the group, the expression of BMP-2 mRNA in the second week was higher than that in the first week (P0.05). Conclusion: ascorbic acid can promote the expression of BMP-2mRNA in osteoblasts.
