分子排阻色譜-多角度激光散射法測(cè)定注射用核糖核酸分子量及其分布

【摘要】:目的測(cè)定注射用核糖核酸中核糖核酸的分子量及其分布。方法采用分子排阻色譜-多角度激光散射聯(lián)用技術(shù),以含0.02%疊氮鈉的0.1 mol·L-1氯化鈉溶液為流動(dòng)相,流速0.5 m L·min-1,采用TSKgel G2000SWXL色譜柱,柱溫為30℃。結(jié)果獲得了樣品的dn/dc值、分子量與分子量分布等分子特征參數(shù)。測(cè)得樣品dn/dc值為0.1547,分子量(Mw)在4.6~234.2 k Da,分子量分布系數(shù)(Mw/Mn)在1.3~2.9。結(jié)論本文嘗試以新的思路建立測(cè)定生物大分子核糖核酸分子量的方法,該方法快速、簡(jiǎn)便,可用于核酸類藥物分子量及其分布的測(cè)定。實(shí)際樣品檢測(cè)結(jié)果表明,市售的注射用核糖核酸的分子量及分布差異均較大,原料及生產(chǎn)工藝有待進(jìn)一步規(guī)范,加強(qiáng)質(zhì)量控制。
