賽庚啶對糖皮質(zhì)激素誘導(dǎo)的大鼠非酒精性脂肪肝的保護(hù)作用研究

【摘要】:目的研究賽庚啶在地塞米松誘導(dǎo)的大鼠非酒精性脂肪肝(NAFLD)中的作用。方法 SD大鼠隨機分為5組,包括正常組、高與低2個劑量模型組(地塞米松2.5,1.0 mg·kg-1)和高與低2個劑量干預(yù)組(賽庚啶5.0,2.0mg·kg-1),實驗共進(jìn)行14 d。實驗結(jié)束后,處死大鼠,稱體重、肝重,計算肝脂數(shù);檢測血清肝功能與血脂指標(biāo)和血糖、游離脂肪酸、胰島素、促腎上腺皮質(zhì)激素及皮質(zhì)酮水平。分離大鼠肝細(xì)胞,加入不同濃度的地塞米松和賽庚啶,油紅O染色觀察肝細(xì)胞內(nèi)脂滴的形成情況,檢測培養(yǎng)上清中糖和細(xì)胞內(nèi)三酰甘油、游離脂肪酸水平。結(jié)果與正常組相比,地塞米松可造成大鼠糖脂代謝紊亂,過度激活下丘腦-垂體-腎上腺皮質(zhì)(HPA)軸,造成促腎上腺皮質(zhì)激素、皮質(zhì)酮水平升高。而賽庚啶可降低HPA軸的反應(yīng)性,改善大鼠的糖脂代謝。在大鼠肝細(xì)胞模型實驗中得到相似的結(jié)果。結(jié)論賽庚啶可通過干預(yù)機體HPA軸,改善地塞米松誘導(dǎo)的大鼠NAFLD。
[Abstract]:Objective to study the effect of cyproheptadine on dexamethasone induced non-alcoholic fatty liver (NAFLD) in rats. Methods Sprague-Dawley rats were randomly divided into 5 groups, including normal group, high and low dose model group (dexamethasone 2.5U 1.0 mg kg-1) and high and low dose intervention groups (cyproheptadine 5.0 mg kg-1) for 14 days. At the end of the experiment, the rats were killed to weigh weight, liver fat number, serum lipids, blood glucose, free fatty acid, insulin, adrenocorticotropic hormone and corticosterone levels. Rat hepatocytes were isolated, dexamethasone and cyproheptadine were added at different concentrations. Lipid droplets in hepatocytes were observed by oil red O staining, and the levels of sugar, triglyceride and free fatty acids in cultured supernatants were detected. Results compared with the normal group, dexamethasone could cause the disorder of glucose and lipid metabolism, over-activate the (HPA) axis of hypothalamus-pituitary-adrenal cortex, and increase the levels of corticotropin and corticosterone. Cyproheptadine could decrease the reactivity of HPA axis and improve the metabolism of glucose and lipid in rats. Similar results were obtained in rat hepatocyte model. Conclusion cyproheptadine can improve the dexamethasone induced NAFLD by interfering with the HPA axis.
