甲基苯丙胺微形態(tài)分析與毒理探討

【摘要】:隨著溶液理論的深入研究,溶劑化學(xué)引起了人們的廣泛關(guān)注。從分子水平研究溶液的性質(zhì),例如離子-離子間締合、離子-分子間溶劑化和溶劑的微觀結(jié)構(gòu)等,對(duì)現(xiàn)代生物化學(xué)發(fā)展具有巨大的指導(dǎo)意義,有助于溶液與生物環(huán)境中控制離子運(yùn)動(dòng)研究的發(fā)展,在溶液分子的空間結(jié)構(gòu)、材料科學(xué)和生命科學(xué)研究等方面也有著重要的作用。本文在總結(jié)前人工作的基礎(chǔ)上,以甲基苯丙胺-單溶劑和雙溶劑溶液體系為研究對(duì)象,采用紅外光譜、熒光光譜技術(shù)并結(jié)合理論計(jì)算方法考察分析了甲基苯丙胺在溶劑體系中的微觀結(jié)構(gòu)變化,討論了其體內(nèi)循環(huán)情況和中毒機(jī)理。主要研究?jī)?nèi)容如下: 1.采用熒光光譜技術(shù)研究了甲基苯丙胺的單溶劑、混合溶劑下的溶液體系的變化情況。通過(guò)對(duì)譜圖中出現(xiàn)溶劑化新峰的分析和計(jì)算,討論了溶液體系中的分子間相互弱作用力對(duì)物質(zhì)微觀形態(tài)的影響。 2.結(jié)合量子化學(xué)計(jì)算得到溶劑化結(jié)構(gòu)的最優(yōu)結(jié)構(gòu)模型,討論了甲基苯丙胺在單溶劑、雙溶劑下微觀形態(tài)的可能構(gòu)型,并通過(guò)分析紅外光譜驗(yàn)證了分子間化學(xué)鍵的變化,由此討論了乙醇對(duì)甲基苯丙胺對(duì)人體循環(huán)的影響。 3.通過(guò)建立甲基苯丙胺誘導(dǎo)小鼠中毒模型,,研究了小鼠在體內(nèi)循環(huán)后的分布情況,得出甲基苯丙胺廣泛擴(kuò)散于人體組織中,尤其以腦部最多。并通過(guò)理論計(jì)算從化學(xué)角度討論了甲基苯丙胺的中毒機(jī)理。
[Abstract]:With the further study of solution theory, solvent chemistry has attracted wide attention. The study of the properties of solution at molecular level, such as ion-ion association, ion-intermolecular solvation and solvent microstructure, is of great significance for the development of modern biochemistry. It is helpful to the research of controlling ion movement in solution and biological environment and plays an important role in the study of spatial structure of solution molecules, material science and life science. In this paper, based on the previous work, the system of methamphetamine-single solvent and double solvent solution was used as the research object, and the infrared spectrum was used. The microstructural changes of methamphetamine in solvent system were investigated and analyzed by fluorescence spectroscopy and theoretical calculation. The internal circulation and poisoning mechanism of methamphetamine were discussed. The main contents are as follows: 1. The change of solution system of methamphetamine in single solvent and mixed solvent was studied by fluorescence spectroscopy. Based on the analysis and calculation of the solvation peak in the spectrum, the influence of the weak interaction between molecules in the solution system on the microstructure of the substance is discussed. 2. Combined with quantum chemical calculation, the optimal structure model of solvated structure was obtained. The possible configuration of the microstructure of methamphetamine in single and double solvents was discussed, and the change of intermolecular chemical bond was verified by IR spectra. The effect of ethanol on the circulation of methamphetamine was discussed. The distribution of methamphetamine in mice was studied by establishing a model of methamphetamine induced poisoning in vivo. It was concluded that methamphetamine spread widely in human tissues, especially in the brain. The mechanism of methamphetamine poisoning was discussed by theoretical calculation.
