HPLC法測定甲磺酸奧希替尼的含量

【摘要】:為了建立一種檢測甲磺酸奧希替尼含量的HPLC法,使用Agilent ZORBAX Eclipse Plus C_(18)色譜柱(4.6 mm×250 mm,5μm),流動相為甲醇-磷酸二氫鈉緩沖液(pH 3.0)(50∶50),檢測波長210 nm,柱溫35℃,流速1.0 mL/min進(jìn)行檢測。甲磺酸奧希替尼濃度在分析物正常濃度的50%~150%(0.201 1~0.603 2 mg/mL)范圍內(nèi)與峰面積呈良好線性關(guān)系(r=0.999 9),最低檢測限為0.08μg/mL,定量限為0.32μg/mL。3批樣品的含量(%)分別為100.1、99.5和99.7。在所建立的色譜條件下,甲磺酸奧希替尼與各有關(guān)物質(zhì)分離良好,包括部分工藝雜質(zhì)及降解雜質(zhì)。結(jié)果表明,所建立的方法專屬、準(zhǔn)確、簡單、耐用,可用于奧希替尼原料的含量測定。
