熱響應(yīng)納米金球作為非共價(jià)藥物載體合成及性質(zhì)研究

【摘要】:以聚N-異丙基丙烯酰胺接枝的聚酰胺作為穩(wěn)定劑,以純水作為溶劑,采用原位合成的方法合成了具有可調(diào)光熱響應(yīng)行為、智能響應(yīng)釋藥行為的有機(jī)無機(jī)雜化金納米粒,并以TEM、紫外-可見分光光度計(jì)測定其在不同溫度下分別對金核的粒徑和表面等離子吸收峰的影響,采用1H-NMR,IR對上述聚合物做了表征.通過紫外-可見光分光光度計(jì)測定了不同組樣品的低臨界溶脹/溶解溫度(LCST)值,發(fā)現(xiàn)不同組樣品LCST有一定差別,通過激光照射測定了樣品的體外光熱響應(yīng)性能.選用難溶性吲哚美辛作為模型藥物,考察了各組樣品的載藥釋藥行為.實(shí)驗(yàn)研究結(jié)果表明,聚N-異丙基丙烯酰胺接枝的聚酰胺修飾的金納米粒較單獨(dú)的聚合物N-聚異丙基丙烯酰胺更適宜作為多功能藥物載體:具有更接近人體生理溫度的LCST值,并且具有隨溫度變化而發(fā)生變化的光熱響應(yīng)性質(zhì)及溫度響應(yīng)的藥物釋放行為.因此它在難溶性藥物的控制釋藥、光熱腫瘤消融領(lǐng)域具有較為廣闊的應(yīng)用前景.
[Abstract]:With polyamide graft of poly (N- isopropyl acrylamide) as a stabilizer and pure water as a solvent, the organic-inorganic hybrid gold nanoparticles with adjustable photothermal response behavior and intelligent response to drug release were synthesized by in situ synthesis, and the particle sizes of gold nuclei at different temperatures were measured with TEM and UV visible spectrophotometer. With the influence of the surface plasmon absorption peak, the above polymer was characterized by 1H-NMR and IR. The low critical swelling / dissolution temperature (LCST) value of different groups of samples was measured by UV visible spectrophotometer. It was found that the different groups of samples were different in LCST, and the photothermal response of the samples was measured by laser irradiation. As a model drug, indomethacin was used as a model drug to investigate the drug release behavior of each sample. The experimental results showed that polyamide modified polyamide grafted N- isopropyl acrylamide was more suitable as a multifunctional drug carrier than N- polyisopropyl acrylamide alone: it had a LCST value closer to the physiological temperature of human body. It has the properties of photothermal response and the drug release behavior of the temperature response. Therefore, it has a broad application prospect in the field of photothermal tumor ablation in the control of drug release from insoluble drugs and in the field of photothermal tumor ablation.
