【摘要】：對(duì)一類肽化合物的抗凝活性進(jìn)行初步篩選,同時(shí)建立測(cè)定大鼠血漿中化合物TI-02濃度的HPLC方法。選用C18柱(250 mm×4.6 mm,10μm),采用甲醇-乙腈-水三元體系梯度洗脫,流速1.0 mL·min~(-1),檢測(cè)波長(zhǎng)240 nm,柱溫30℃,進(jìn)樣量20μL�？鼓衔颰I-02在濃度0.125~10.0μg/mL范圍內(nèi)線性良好,r=0.9964;回收率為97.1%~106.7%;日內(nèi)、日間精密度RSD2.3%,室溫24 h內(nèi)穩(wěn)定,RSD1.0%。本實(shí)驗(yàn)建立HPLC測(cè)定方法靈敏度高、精密度好,結(jié)果穩(wěn)定可靠,可用于該肽類化合物大鼠血漿中濃度的測(cè)定。
