載氯諾昔康殼聚糖關(guān)節(jié)腔注射用溫敏凝膠的制備及體外釋藥性觀察

【摘要】:目的制備關(guān)節(jié)腔注射用載氯諾昔康(Lnxc)殼聚糖溫敏凝膠,觀察其理化性質(zhì)及體外釋藥性。方法以殼聚糖、β-甘油磷酸鈉(β-GP)為材料制備載Lnxc溫敏凝膠,以膠凝溫度、時間、通針性為評價指標(biāo),優(yōu)化處方工藝;采用椎板法觀察凝膠的流變學(xué)性質(zhì),采用離心試驗、溫度試驗觀察凝膠的穩(wěn)定性,采用動態(tài)膜透析法觀察載Lnxc溫敏凝膠和Lnxc溶液的體外釋藥特性。結(jié)果載Lnxc殼聚糖溫敏凝膠的最佳處方為Lnxc 2 mg、3%殼聚糖、60%β-GP、p H 7.2,所制備的溫敏凝膠屬于非牛頓流體,離心后無分層現(xiàn)象,4℃和25℃條件下呈流動狀,37℃發(fā)生相變形成凝膠。Lnxc溶液8 h藥物累積釋放率86.70%,載Lnxc溫敏凝膠72 h藥物累積釋放率86.41%,釋藥規(guī)律符合Higuchi方程。結(jié)論成功制備載Lnxc殼聚糖關(guān)節(jié)腔用溫敏凝膠,其具有理想的膠凝溫度和較好的緩釋效果。
[Abstract]:Objective to prepare lornoxicam (Lnxc) chitosan thermosensitive gel for intraarticular injection and observe its physicochemical properties and drug release in vitro. Methods Lnxc thermosensitive gel was prepared by using chitosan and 尾 -GP as materials, the gelation temperature, time and needle permeability were used as evaluation indexes to optimize the prescription process, the rheological properties of the gel were observed by laminar method, and the centrifugal test was used. The stability of the gel was observed by temperature test, and the in vitro release characteristics of Lnxc thermo-sensitive gel and Lnxc solution were observed by dynamic membrane dialysis. Results the best formulation of Lnxc chitosan temperature-sensitive gel was: Lnxc 2 mg / g 3% chitosan 60% 尾 -GPP H 7.2. The temperature sensitive gel was a non-Newtonian fluid.
