1-甲基乙內(nèi)酰脲皮下注射及胃飼降低大鼠生長(zhǎng)激素水平的實(shí)驗(yàn)

【摘要】:目的: 探索1-甲基乙內(nèi)酰脲不同給藥途徑、給藥劑量對(duì)降低大鼠生長(zhǎng)激素的影響;探究胃飼是否同樣具有降低生長(zhǎng)激素的作用。 方法: 實(shí)驗(yàn)對(duì)象:成年雌性SD大鼠80只。分組:將80只雌性SD大鼠按給藥途徑不同隨機(jī)分為皮下注射組(H1-H5)和胃飼組(G1-G5)兩個(gè)大組、10個(gè)小組,每小組大鼠8只;每個(gè)大組設(shè)空白對(duì)照組(H1組、G1組)、造模對(duì)照組(H2組、G2組)、實(shí)驗(yàn)組(H3-H5組和G3-G5組)。處理:空白對(duì)照組(H1組、G1組)予以生理鹽水0.5ml腹腔注射。造模對(duì)照組(H2組、G2組)予以10mg/ml苯巴比妥0.5ml腹腔注射造模。實(shí)驗(yàn)組先予以10mg/ml苯巴比妥0.5ml腹腔注射造模,造模后10分鐘再對(duì)各個(gè)實(shí)驗(yàn)組按照不同給藥途徑及劑量給藥:H3組、G3組給予1-甲基乙內(nèi)酰脲0.1mg,H4組、G4組給予1-甲基乙內(nèi)酰脲1.0mg,H5組、G5組給予1-甲基乙內(nèi)酰脲10mg);空白對(duì)照組(H1組、G1組)、造模對(duì)照組(H2組、G2組)、造模后10分鐘,則分別給予PBS0.5ml。取樣:各組均在注射生理鹽水或苯巴比妥造模后20分鐘、30分鐘、40分鐘時(shí)由尾靜脈取血,離心取上層血清。檢測(cè):按大鼠生長(zhǎng)激素試劑盒說明做血清ELISA檢測(cè)生長(zhǎng)激素水平,對(duì)比各組之間生長(zhǎng)激素水平變化。結(jié)果分析:應(yīng)用計(jì)算軟件SPSS18.0,統(tǒng)計(jì)分析得出結(jié)果。 結(jié)果: 1.造模對(duì)照組較空白對(duì)照組大鼠生長(zhǎng)激素水平增高顯著,空白對(duì)照組腹腔注射生理鹽水后30分鐘生長(zhǎng)激素水平為5.022±0.469ug/L。造模對(duì)照組的造模后30分鐘生長(zhǎng)激素水平為10.00±0.748ug/L,兩組生長(zhǎng)激素值差異有統(tǒng)計(jì)學(xué)意義(P0.05),提示造模成功。 2.皮下注射組可以有效降低大鼠生長(zhǎng)激素的水平。皮下注射1-甲基乙內(nèi)酰脲10mg組20分鐘(即造模后30分鐘),生長(zhǎng)激素水平為2.960±0.202ug/L。皮下注射1-甲基乙內(nèi)酰脲1mg組及0.1mg組均可降低大鼠生長(zhǎng)激素的水平,與造模對(duì)照組比較生長(zhǎng)激素顯著降低,差異均具有統(tǒng)計(jì)學(xué)意義(P0.05),提示皮下注射1-甲基乙內(nèi)酰脲可顯著降低大鼠生長(zhǎng)激素的水平。 3.胃飼組可有效降低大鼠生長(zhǎng)激素的水平;胃飼1-甲基乙內(nèi)酰脲10mg/只組30分鐘(即造模后40分鐘),生長(zhǎng)激素水平為4.212±0.525ug/L。胃飼1-甲基乙內(nèi)酰脲1mg組及0.1mg組均可降低大鼠生長(zhǎng)激素的水平,,與造模對(duì)照組比較生長(zhǎng)激素顯著降低,差異均具有統(tǒng)計(jì)學(xué)意義(P0.05),提示胃飼1-甲基乙內(nèi)酰脲可顯著降低大鼠生長(zhǎng)激素的水平。 4.皮下注射效果要優(yōu)于胃飼組皮下注射組:皮下注射1-甲基乙內(nèi)酰脲10mg降低大鼠生長(zhǎng)激素的最低值2.960±0.202ug/L,胃飼1-甲基乙內(nèi)酰脲10mg降低大鼠生長(zhǎng)激素最低值4.212±0.525ug/L,兩組生長(zhǎng)激素值差異有統(tǒng)計(jì)學(xué)意義(P0.05),提示皮下注射1-甲基乙內(nèi)酰脲優(yōu)于胃飼。 5.皮下注射組降低大鼠生長(zhǎng)激素的最低值在用藥后20分鐘(即造模后30分鐘),而胃飼組在用藥后30分鐘(即造模后40分鐘)時(shí)間段的大鼠生長(zhǎng)激素的下降最明顯。 結(jié)論: 1.皮下注射及胃飼1-甲基乙內(nèi)酰脲均具有降低大鼠生長(zhǎng)激素水平的作用; 2.皮下注射1-甲基乙內(nèi)酰脲降低大鼠生長(zhǎng)激素效果優(yōu)于胃飼;
[Abstract]:Objective:
To explore the different routes of administration of 1- methylhydantoin and the effect of dosage on the growth hormone of rats; explore whether gastric feeding also has the effect of reducing growth hormone.
Method:
Subjects: 80 adult female SD rats were divided into groups: 80 female SD rats were randomly divided into subcutaneous injection group (H1-H5) and stomach feeding group (G1-G5) in two large groups, 10 groups and 8 rats in each group; each group had blank control group (group H1, G1), model control group (H2 group, G2 group), experimental group (H3-H5 group and G3-G5 group). Treatment: the blank control group (group H1, group G1) was given 0.5ml intraperitoneal injection of saline 0.5ml. The model control group (group H2, G2) was injected with 10mg/ml phenobarbital 0.5ml intraperitoneal injection molding. The level of growth hormone was detected by the rat growth hormone kit, and the level of growth hormone was compared with the growth hormone level between each group. The results were analyzed by the application of computing software SPSS18.0, and the results were obtained by statistical analysis.
Result:
1. the growth hormone level in the control group was significantly higher than that in the blank control group. The growth hormone level of the control group was 10 + 0.748ug/L after the 30 minute model of the control group with the growth hormone level of 5.022 + 0.469ug/L. after intraperitoneal injection of saline (P0.05), and the difference of growth hormone values in the two groups was statistically significant (P0.05). Success.
2. subcutaneous injection group can effectively reduce the level of growth hormone in rats. Subcutaneous injection of 1- methylhydantoin 10mg group 20 minutes (i.e. 30 minutes after modeling), growth hormone level of 2.960 + 0.202ug/L. subcutaneous injection of 1- methyl hydanreon 1mg group and 0.1mg group can reduce the level of rat growth irritable, compared with the model control group, the growth of Ji Suxian was compared with the model control group. The difference was statistically significant (P0.05), suggesting that subcutaneous injection of 1- methylhydantoin could significantly reduce the level of growth hormone in rats.
3. gastric feeding group can effectively reduce the level of growth hormone in rats; gastric feeding 1- methylhydantoin 10mg/ is only 30 minutes (i.e. 40 minutes after modeling), growth hormone level is 4.212 + 0.525ug/L. gastric feeding 1- methyl hydantoin 1mg group and 0.1mg group can reduce the level of growth hormone in rats, compared with the model control group, the growth hormone is significantly lower than that in the model control group. 4. the effect of subcutaneous injection was better than that in the subcutaneous injection group of the stomach feeding group: the minimum value of 1- methylhydantoin 10mg subcutaneous injection was 2.960 + 0.202ug/L, and the lowest growth hormone of the gastric feeding 1- methylhydantoin 10mg was reduced by 4.212 + 0.525ug/L, and the difference of growth hormone value was statistically significant (P0.05), suggesting subcutaneous injection of 1-. Methylhydantoin is superior to gastric feeding.
5. the lowest value of the growth hormone in the subcutaneous injection group was 20 minutes after the medication (30 minutes after the model was made), while the growth hormone of the rats in the stomach feeding group was the most obvious at 30 minutes after the medication (that is, 40 minutes after the model 40 minutes).
Conclusion:
1. subcutaneous injection and gastric feeding of 1- methylhydantoin can reduce the growth hormone level in rats.
2. the effect of subcutaneous injection of 1- methylhydantoin on growth hormone in rats was better than that in gastric feeding.
