【摘要】：由于生物制品具有復雜的特征和性質(zhì),為評估申請者的生物相似性證據(jù),美國FDA建議使用"基于風險"和"證據(jù)鏈完備性"(totality-of-the-evidence)的方式來評估已遞交的全部特征域間的所有可用數(shù)據(jù)和信息。因此,需要建立一個生物類似性指數(shù),使該指數(shù)能將各個具體特征域中的生物相似性整合成為一個跨指標領域的總體指數(shù)(global index)。Shein-Chung Chow等人已經(jīng)在2010年根據(jù)受試品-參考品(T-R)與參考品-參考品(R-R)比較研究中重現(xiàn)性概率(reproducibility probability)的概念,提出了一個評估相似性通用的方法,該方法可用于"證據(jù)鏈完備性"的評估。本文提出另外一個生物相似性評價指數(shù),該指數(shù)基于配對隨機設計下隨機系數(shù)的線性回歸模型,并提供了相應的統(tǒng)計檢測步驟。文中用一個數(shù)值型實例對所提出的方法進行了證明。
[Abstract]:Because of the complex characteristics and nature of biological products, in order to assess the applicant's biological similarity evidence, The FDA recommends the use of "risk-based" and "evidence-chain completeness" (totality-of-the-evidence) methods to evaluate all available data and information between all submitted feature domains. So there is a need for a biological similarity index, (global index). Shein-Chung Chow et al., which enables the index to integrate biological similarity across specific feature domains into a cross-index domain, have been based on a comparative study of test-take-reference (T-R) and reference-reference (R-R) studies in 2010. The concept of reproducibility probability (reproducibility probability), A general method for evaluating similarity is presented, which can be used to evaluate the completeness of evidence chain. In this paper, another biological similarity evaluation index is proposed, which is based on the linear regression model of random coefficients under pairwise random design, and provides the corresponding statistical detection steps. The proposed method is proved by a numerical example.
