【摘要】：安舒法辛是ODV（ODV）的酯化前藥，在體內(nèi)經(jīng)酯酶水解后轉(zhuǎn)化為活性產(chǎn)物ODV，屬于5-羥色胺-去甲腎上腺素重?cái)z取抑制劑（SNRI）型抗抑郁藥，具有組織突出前細(xì)胞受體作用功能，致使體液中5-羥色胺和去甲腎上腺素濃度升高的藥理活性。本研究建立了適用于測(cè)定Wistar大鼠血漿、腦組織以及下丘腦組織中安舒法辛和ODV含量的LC-MS/MS分析方法，以此對(duì)鹽酸安舒法辛在大鼠體內(nèi)的吸收藥動(dòng)學(xué)和腦組織分布特征進(jìn)行了考察，并與ODV的此方面特征進(jìn)行了對(duì)比和分析。利用DAS3.0、SPSS13.0等軟件對(duì)藥代參數(shù)進(jìn)行了計(jì)算并對(duì)結(jié)果進(jìn)行了統(tǒng)計(jì)學(xué)分析；同時(shí)對(duì)兩種藥物在體內(nèi)靶向位點(diǎn)—腦部的分布情況進(jìn)行了比較和分析。本研究開發(fā)了快速、靈敏、準(zhǔn)確的分析方法，，為安舒法辛在臨床上的應(yīng)用提供了必要的參考數(shù)據(jù)。
[Abstract]:Amsufaxine is a pre-esterification drug of ODV, which is transformed into the active product ODV by esterase hydrolysis in vivo. It belongs to serotonin norepinephrine reuptake inhibitor (SNRI) type antidepressant, and has the function of tissue protruding precellular receptor. The pharmacological activity that results in increased concentrations of serotonin and norepinephrine in body fluids. In this study, a LC-MS / MS analysis method was established for the determination of ambrofacin and ODV in plasma, brain and hypothalamus of Wistar rats. The absorption pharmacokinetics and brain tissue distribution of amsufacin hydrochloride in rats were investigated. And the characteristics of ODV are compared and analyzed. The pharmacokinetic parameters were calculated by DAS3.0 and SPSS 13.0 software, and the results were statistically analyzed, and the distribution of the two drugs in the brain targeting sites in vivo was compared and analyzed. In this study, a rapid, sensitive and accurate analytical method has been developed, which provides the necessary reference data for the clinical application of ambrofacin.
