超高效液相色譜質(zhì)譜聯(lián)用法測定兒童血漿中利福平、利福噴丁和利福布汀的濃度研究

【摘要】:目的建立超高效液相色譜質(zhì)譜聯(lián)用法(UPLC-MS/MS)同時測定兒童血漿中利福平、利福噴丁和利福布汀的濃度方法。方法以扎來普隆為內(nèi)標(biāo),血漿樣品經(jīng)乙腈沉淀蛋白處理后檢測。ACQUITY UPLC HSS T3色譜柱(2.1 mm×100 mm,1.8μm)為分析柱,以乙腈-水(15 mmol·L-1甲酸銨-0.05%甲酸)為流動相,梯度洗脫,用電噴霧離子化源(ESI),以正離子多反應(yīng)監(jiān)測(MRM),檢測專屬性、標(biāo)準(zhǔn)曲線與定量下限、精密度與回收率、基質(zhì)效應(yīng)和穩(wěn)定性。結(jié)果利福平、利福噴丁和利福布汀的線性范圍分別為100~5000,20~2000,20~2000 ng·m L-1。日內(nèi)、日間精密度相對標(biāo)準(zhǔn)偏差(RSD)均小于15%。結(jié)論本法靈敏、簡便、準(zhǔn)確,可同時對利福平、利福噴丁及利福布汀進(jìn)行治療藥物監(jiān)測。
[Abstract]:Objective to establish a high performance liquid chromatography-mass spectrometry (UPLC-MS / MS) method for simultaneous determination of rifampicin, rifampicin and rifambutin in children's plasma. Methods Zaleplon was used as the internal standard, the plasma sample was detected by acetonitrile precipitation protein treatment. The chromatographic column (2.1 mm 脳 100mm ~ 1.8 渭 m) of UPLC HSS in plasma was used as the analytical column, and acetonitrile-water (15 mmol L-1 ammonium formate -0.05% formic acid) was used as the mobile phase, and the gradient elution was performed. Electrospray ionization source (ESI), positive ion multireaction monitoring (MRM), detection specificity, standard curve and quantitative lower limit, precision and recovery, matrix effect and stability. Results the linear range of rifampicin, rifapentine and rifambutin was 100 ~ 5000m ~ (-1) ~ 20 ~ 200ng / L ~ (-1). Within days, the relative standard deviation (RSD) of inter-day precision was less than 15. Conclusion this method is sensitive, simple and accurate, and can be used to monitor rifampicin, rifampicin and rifambutin simultaneously.
