離子色譜快速檢測殼寡糖含量

【摘要】:目的:為了快速準確的進行殼寡糖樣品的檢測,建立一種分離度好、靈敏度高的殼寡糖離子色譜快速檢測方法。方法:實驗選用Dionex-Carbo Pac PA104×50 mm離子交換色譜柱,通過優(yōu)化柱溫、淋洗液配比及濃度等色譜條件,將不同聚合度的殼寡糖在30 min內(nèi)實現(xiàn)分離。分離后的殼寡糖樣品通過計算聚合度為殼2~6糖的含量,檢驗其樣品中殼寡糖的純度。結(jié)果:樣品濃度在2.00~10.00μg·m L~(-1)范圍內(nèi)線性關系良好(r≥0.999 0),精密度實驗RSD在0.1%~1.6%之間,樣品加標回收率93.2%~105.0%。3種殼寡糖樣品中2~6個聚合度殼寡糖的含量之和分別為25.944μg·g~(-1)、26.406μg·g~(-1)、25.374μg·g~(-1),計算得到純度為86.5%、88.0%和84.6%,符合國家標準要求。結(jié)論:該方法需要樣品量少,檢驗快速,無需衍生,為殼寡糖質(zhì)量分析、組分分離提供了依據(jù)。
[Abstract]:Objective: to establish a rapid and sensitive method for the determination of chitosan oligosaccharides by ion chromatography. Methods: Dionex-Carbo PA104 脳 50mm ion exchange column was used to separate chitosan oligosaccharides with different degree of polymerization within 30 min by optimizing the column temperature, eluant ratio and concentration.
