RGD肽修飾的異長(zhǎng)春花堿-粉防己堿脂質(zhì)體的制備及體外抑瘤作用研究

【摘要】:目的:制備RGD肽修飾的異長(zhǎng)春花堿(VRB)-粉防己堿(TET)脂質(zhì)體,研究其對(duì)腦膠質(zhì)瘤C6細(xì)胞的抑制作用。方法:采用薄膜分散法和硫酸銨梯度法制備RGD肽修飾的VRB-TET脂質(zhì)體,觀察其形態(tài)和粒徑分布,測(cè)定其中VRB的含量;以磺酰羅丹明B法分別測(cè)定空白靶向脂質(zhì)體、VRB普通脂質(zhì)體和RGD肽修飾的VRB-TET脂質(zhì)體對(duì)C6細(xì)胞的抑制作用。結(jié)果:所制RGD肽修飾的VRB-TET脂質(zhì)體呈圓球狀或類(lèi)圓球狀,表面光滑,粒徑為120 nm左右,其中VRB的平均含量為28.27μg/m L(RSD=0.38%,n=3)。空白靶向脂質(zhì)體對(duì)C6細(xì)胞生長(zhǎng)無(wú)顯著影響;RGD肽修飾的VRB-TET脂質(zhì)體能明顯抑制C6細(xì)胞生長(zhǎng),其作用后細(xì)胞存活率明顯低于VRB普通脂質(zhì)體(P0.05)。結(jié)論:成功制得RGD肽修飾的VRB-TET脂質(zhì)體,其具有明顯的抑制C6細(xì)胞生長(zhǎng)的作用。
[Abstract]:Aim: to prepare RGD peptide modified liposomes of VRB- Tetrandrine (Tetrandrine) and study its inhibitory effect on glioma C6 cells. Methods: RGD peptide modified VRB-TET liposomes were prepared by thin-film dispersion and ammonium sulfate gradient method. The morphology and particle size distribution of VRB-TET liposomes were observed and the content of VRB was determined. The inhibitory effects of VRB liposomes and VRB-TET liposomes modified with RGD peptide on C6 cells were determined by sulfonyl rhodamine B method. Results: the RGD peptide modified VRB-TET liposomes were spherical or spherical in shape with smooth surface and about 120 nm diameter. The average content of VRB was 28.27 渭 g / mL RSD0.38. Blank targeted liposomes had no significant effect on the growth of C6 cells. The RGD peptide-modified VRB-TET liposomes could significantly inhibit the growth of C6 cells, and the cell survival rate was significantly lower than that of VRB liposome P0.05. Conclusion: VRB-TET liposomes modified by RGD peptide can inhibit the growth of C6 cells.
