HepG2.2.15細(xì)胞的差異性表達(dá)及對(duì)IFN-α的反應(yīng)性研究

【摘要】:采用有限稀釋法從HepG2.2.15細(xì)胞株中分離出10株單克隆細(xì)胞株。ELISA法檢測(cè)其HBsAg與HBe Ag表達(dá)量,根據(jù)HBsAg、HBe Ag表達(dá)量的不同,篩選出最高、中等、最低3株克隆細(xì)胞株。MTT法進(jìn)行干擾素(IFN)-α的細(xì)胞毒作用研究,表明IFN-α對(duì)不同克隆細(xì)胞株藥物毒作用相似。ELISA法檢測(cè)各克隆細(xì)胞株給IFN-α前后HBsAg、HBe Ag表達(dá)量,IFN-α對(duì)HepG2.2.15細(xì)胞HBe Ag的抑制作用較弱,對(duì)HBsAg、HBV DNA的抑制作用較強(qiáng),且對(duì)表達(dá)量高的克隆細(xì)胞株抑制作用更強(qiáng)。不同HepG2.2.15細(xì)胞表達(dá)HBsAg與HBe Ag存在差異性,且表達(dá)量高的細(xì)胞株對(duì)治療濃度的IFN-α更加敏感。
[Abstract]:The expression of HBsAg and HBe Ag in HepG2.2.15 cell line was detected by Elisa method. The HBsAg and HBe Ag expression levels were determined by using the limited dilution method. According to the different expression levels of HBe Ag in HBsAg, the highest and moderate HBe Ag expression levels were obtained from HepG2.2.15 cell line. The cytotoxicity of IFN- 偽 was studied by MTT assay. The results showed that IFN- 偽 had similar drug toxicity to different clone cell lines. Elisa method was used to detect the expression of HBsAg HBe Ag in HBe cells before and after IFN- 偽 was given. IFN- 偽 had a weak inhibitory effect on HBe Ag in HepG2.2.15 cells, and a stronger inhibitory effect on HBe DNA in HBN- 偽 cells before and after IFN- 偽 administration. Moreover, it had stronger inhibitory effect on clone cell lines with high expression. Different HepG2.2.15 cells expressed HBsAg and HBe Ag differently, and the cell lines with high expression were more sensitive to IFN- 偽.
