【摘要】：探討沒食子酸與環(huán)丙沙星聯(lián)用對小鼠慢性鼻-鼻竇炎模型(CRS)的作用。收集難治性CRS患者鼻黏膜樣本,分離銅綠假單胞桿菌(Pseudomonas aeruginosa)構(gòu)建小鼠CRS模型,沒食子酸(GA)與環(huán)丙沙星(CIP)單獨(dú)或聯(lián)合灌胃治療,HE染色觀察小鼠鼻黏膜病理變化,ELISA檢測血清炎性因子TNF-α、IL-6、IL-8的表達(dá)、試劑盒檢測SOD活性、MDA及ROS含量,Western blot檢測鼻黏膜組織中TNF-α、IL-6、IL-8、IκB及NF-κB p65的表達(dá)。結(jié)果顯示,GA與CIP可顯著降低CRS小鼠TNF-α、IL-6、IL-8、MDA、ROS及NF-κB p65的含量,增加SOD活性及IκB的表達(dá),且聯(lián)合用藥效果更顯著。結(jié)果說明,GA與CIP聯(lián)用治療CRS效果優(yōu)于單獨(dú)用藥,可能通過抑制NF-κB信號通路,下調(diào)TNF-α、IL-6、IL-8表達(dá)發(fā)揮作用。
[Abstract]:To investigate the effect of Gallic acid combined with ciprofloxacin on chronic rhinosinusitis in mice. A mouse CRS model was established by collecting nasal mucosa samples from refractory CRS patients and isolating Pseudomonas aeruginosa. The expression of serum inflammatory factor TNF- 偽, IL-6 and IL-8 was detected by Elisa in the nasal mucosa of mice treated with HE alone or in combination with ciprofloxacin (CIP), Gallic acid (GAG) and ciprofloxacin (ciprofloxacin). The activity of SOD and the content of Ros in nasal mucosa were detected by Western blot. The expression of TNF- 偽, IL-6, IL-8, I 魏 B and NF- 魏 B p65 in nasal mucosa was detected. The results showed that GA and CIP could significantly decrease the contents of TNF- 偽 IL-6, IL-8, MDAROS and NF- 魏 B p65, increase the activity of SOD and the expression of I 魏 B in CRS mice. The results showed that the combination of TNF- 偽 and CIP in the treatment of CRS was better than that in the treatment of CRS alone, which might play a role by inhibiting the signal pathway of NF- 魏 B and down-regulating the expression of TNF- 偽 -IL-6 / IL-8.
