大鼠血漿中7-乙基-10-羥基喜樹堿-10-棕櫚酸酯及7-乙基-10-羥基喜樹堿HPLC分析方法的建立

【摘要】:目的建立同時測定大鼠血漿中7-乙基-10-羥基喜樹堿-10-棕櫚酸酯(SN38-PA)及其活性代謝產(chǎn)物7-乙基-10-羥基喜樹堿(SN38)含量的高效液相色譜法。方法以10-羥基喜樹堿(10-CPT)作為內(nèi)標,甲酸酸化血漿,甲醇沉淀蛋白。采用Agilent C18柱(4.6 mm×250 mm,5μm)色譜柱,以甲醇-0.2%甲酸水溶液為流動相進行梯度洗脫,流速1 ml/min,紫外檢測波長372 nm,柱溫:30℃。結(jié)果大鼠血漿中SN38-PA和SN38線性范圍分別為0.25~62.5(r=0.9998)和0.05~12.5μg/ml(r=0.9997),最低定量限分別為0.18和0.04μg/ml;兩組分平均絕對回收率分別為95.89%和97.03%;平均相對回收率分別為99.54%和99.84%。2組分日內(nèi)、日間精密度的RSD%均3%。結(jié)論本分析方法快速、簡便、準確度和靈敏度高,可用于SN38-PA及其活性代謝產(chǎn)物SN38的體內(nèi)檢測。
[Abstract]:Objective to establish a HPLC method for simultaneous determination of 7-ethyl-10-hydroxycamptothecin (7-ethyl-10-hydroxycamptothecin) and its active metabolite (7-ethyl-10-hydroxycamptothecin) in rat plasma. Methods 10-hydroxycamptothecin-10-CPT was used as internal standard. Plasma was acidified with formic acid and protein was precipitated by methanol. A Agilent C18 column (4.6 mm 脳 250 mm ~ 5 渭 m) was used for gradient elution with methanol-0.2% formic acid solution as mobile phase.
