替米沙坦對兔髂動脈再狹窄模型新生內(nèi)膜增殖的影響

【摘要】:目的觀察兔髂動脈再狹窄模型病理改變,并加用替米沙坦進行干預(yù),探討其對再狹窄模型新生內(nèi)膜增殖的影響。方法 30只新西蘭大白兔隨機分為對照、再狹窄和替米沙坦組,每組10只。采用高脂飼料喂養(yǎng)聯(lián)合兔髂動脈二次球囊損傷建立再狹窄模型,其中替米沙坦組于第二次球囊損傷后第2天開始加用替米沙坦5mg/(kg·d)。實驗結(jié)束時,動物處死前采血檢測總膽固醇,并留取髂動脈標本,行蘇木精伊紅染色法染色及免疫組化檢測,組織勻漿測定血管緊張素Ⅱ(AngⅡ)水平。結(jié)果與對照組比較,再狹窄和替米沙坦組總膽固醇[(20.44±0.68)、(20.39±0.60)比(0.58±0.19)mmol/L]、血管壁AngⅡ[(115.6±15.7)、(123.8±22.1)比(90.1±7.7)ng/L]、內(nèi)膜厚度[(266.1±70.3)、(68.2±24.4)比(2.8±0.2)μm]和狹窄率[(89.3±6.9)%、(42.6±18.1)%比(23.0±3.5)%]增加(均P0.05)。而替米沙坦組的內(nèi)膜厚度和狹窄率低于再狹窄組(均P0.01)。與對照和替米沙坦組比較,再狹窄組平滑肌細胞增殖率明顯增加[(21.9±4.7)%比(7.3±4.9)%、(7.1±5.1)%,均P0.01]。α平滑肌肌動蛋白染色證實,再狹窄和替米沙坦組的新生內(nèi)膜主要細胞成分為血管平滑肌細胞。結(jié)論替米沙坦對兔髂動脈再狹窄模型血管平滑肌細胞增殖及新生內(nèi)膜增殖均有抑制作用。
[Abstract]:Objective to observe the pathological changes of rabbit iliac artery restenosis model and to investigate the effect of telmisartan on neointimal proliferation in restenosis model. Methods Thirty New Zealand rabbits were randomly divided into control group, restenosis group and telmisartan group with 10 rabbits in each group. The restenosis model of iliac artery was established by high fat diet feeding combined with secondary balloon injury of iliac artery in rabbits. Telmisartan 5mg/(kg dronium was added to telmisartan group on the second day after the second balloon injury. At the end of the experiment, the blood samples were collected to detect total cholesterol before the animals were killed, and the iliac artery specimens were collected.
