【摘要】：目的建立同時測定人血漿中苯海拉明、氯苯那敏、異丙嗪、氯雷他定、非索非那定、地氯雷他定、左西替利嗪、賽庚啶濃度的超高效液相色譜質(zhì)譜(UPLC-MS/MS)法。方法以鹽酸吡格列酮為內(nèi)標,血漿經(jīng)甲醇直接沉淀后進樣分析,用UPLC-MS/MS法進行檢測。色譜柱為ACQUITY UPLC HSS C18柱,流動相為含0.01%甲酸的水溶液-0.01%甲酸的甲醇溶液,流速為0.3 m L·min 230001,China,電噴霧離子源,用多反應(yīng)監(jiān)測,正離子分段掃描分析。結(jié)果苯海拉明、氯苯那敏、異丙嗪、氯雷他定、非索非那定、地氯雷他定、左西替利嗪、賽庚啶血藥濃度分別在1.15×10~(-3)~2.30×10~(-1),2.48×10~(-4)~2.48×10~(-2),2.86×10~(-3)~2.86×10~(-1),2.50×10~(-4)~2.50×10~(-2),5.50×10~(-3)~2.20,2.20×10~(-4)~2.20×10~(-2),5.70×10~(-3)~2.28,3.50×10~(-3)~3.50×10~(-1)μg·m L~(-1)線性關(guān)系良好;最低定量下限分別為1.15×10~(-3),2.48×10~(-4),2.86×10~(-3),2.50×10~(-4),5.50×10~(-3),2.20×10~(-4),5.70×10~(-3),3.50×10~(-3)μg·m L~(-1)。日內(nèi)、日間精密度RSD均15%;提取回收率均65%,RSD均15%。結(jié)論該方法靈敏、快速、準確、專屬性強,可用于上述8種抗組胺類藥物的血藥濃度監(jiān)測及藥代動力學(xué)研究。
[Abstract]:Objective to establish a method for the simultaneous determination of phenhydramine, chlorphenamine, promethazine, loratadine, fexofenadine, desloratadine, levcetirizine and cyproheptadine in human plasma by UPLC-MS / MS. Methods pioglitazone hydrochloride was used as internal standard, plasma was analyzed by direct precipitation of methanol, and detected by UPLC-MS / MS method.
