梔子苷聯(lián)合美滿霉素拮抗PC12細(xì)胞凋亡的作用

【摘要】:目的:觀察梔子苷和美滿霉素聯(lián)合應(yīng)用對(duì)H_2O_2誘導(dǎo)PC12細(xì)胞凋亡的作用,并對(duì)其機(jī)制進(jìn)行初步探討。方法:處于對(duì)數(shù)生長(zhǎng)期的PC12細(xì)胞隨機(jī)分為對(duì)照組、模型組、梔子苷組、美滿霉素組以及梔子苷/美滿霉素聯(lián)合應(yīng)用組。用300μmol/L H_2O_2誘導(dǎo)6 h建立PC12細(xì)胞凋亡模型。MTT法測(cè)定細(xì)胞存活率,比色法測(cè)定乳酸脫氫酶(LDH)活性,倒置相差顯微鏡觀察細(xì)胞形態(tài),Hoechest 33258熒光核染色觀察細(xì)胞凋亡狀況,Annexin V-FITC/PI雙染法流式細(xì)胞儀檢測(cè)細(xì)胞凋亡率,Western Blot測(cè)定Caspase-3蛋白表達(dá)。結(jié)果:與模型組相比,梔子苷組和美滿霉素組PC12細(xì)胞存活率提高,LDH釋放量減少,細(xì)胞凋亡率降低,同時(shí)Caspase-3蛋白表達(dá)減少(P均0.05);與梔子苷組和美滿霉素組相比,梔子苷/美滿霉素聯(lián)合應(yīng)用組PC12細(xì)胞的存活率進(jìn)一步提高,LDH釋放量進(jìn)一步減少,細(xì)胞凋亡率進(jìn)一步降低,同時(shí)Caspase-3蛋白表達(dá)進(jìn)一步減少(P0.05或P0.01)。以上指標(biāo)相比,差異均有統(tǒng)計(jì)學(xué)意義。結(jié)論:梔子苷與美滿霉素聯(lián)合應(yīng)用較單一應(yīng)用能更有效拮抗PC12細(xì)胞凋亡,其機(jī)制可能與抑制Caspase-3蛋白表達(dá)有關(guān)。
[Abstract]:Aim: to observe the effect of geniposide combined with minocycline on apoptosis of PC12 cells induced by H_2O_2 and its mechanism. Methods: PC12 cells at logarithmic growth stage were randomly divided into control group, model group, geniposide group, minocycline group and combined application of geniposide and minocycline. The apoptosis model of PC12 cells induced by 300 渭 mol/L H_2O_2 for 6 h was established. The survival rate of PC12 cells was measured by MTT method, and the activity of lactate dehydrogenase (LDH) was measured by colorimetric method. Apoptosis was observed by inverted phase contrast microscope and fluorescence nuclear staining of Hoechest 33258. The apoptosis rate was detected by flow cytometry with Annexin V-FITC/PI double staining and the expression of Caspase-3 protein was detected by Western Blot. Results: compared with the model group, the survival rate of PC12 cells in geniposide group and minocycline group was decreased, the cell apoptosis rate was decreased, and the expression of Caspase-3 protein was decreased (P < 0.05), compared with those in geniposide group and minocycline group. In combination with geniposide / minocycline group, the survival rate of PC12 cells was further increased, the cell apoptosis rate was further decreased, and the expression of Caspase-3 protein was further decreased by P0.05 or P0.01. Compared with the above indexes, the differences were statistically significant. Conclusion: the combination of geniposide and minocycline can antagonize the apoptosis of PC12 cells more effectively than that of minocycline alone, and the mechanism may be related to the inhibition of Caspase-3 protein expression.
