【摘要】：目的:觀察梔子苷和美滿霉素聯(lián)合應用對H_2O_2誘導PC12細胞凋亡的作用,并對其機制進行初步探討。方法:處于對數(shù)生長期的PC12細胞隨機分為對照組、模型組、梔子苷組、美滿霉素組以及梔子苷/美滿霉素聯(lián)合應用組。用300μmol/L H_2O_2誘導6 h建立PC12細胞凋亡模型。MTT法測定細胞存活率,比色法測定乳酸脫氫酶(LDH)活性,倒置相差顯微鏡觀察細胞形態(tài),Hoechest 33258熒光核染色觀察細胞凋亡狀況,Annexin V-FITC/PI雙染法流式細胞儀檢測細胞凋亡率,Western Blot測定Caspase-3蛋白表達。結(jié)果:與模型組相比,梔子苷組和美滿霉素組PC12細胞存活率提高,LDH釋放量減少,細胞凋亡率降低,同時Caspase-3蛋白表達減少(P均0.05);與梔子苷組和美滿霉素組相比,梔子苷/美滿霉素聯(lián)合應用組PC12細胞的存活率進一步提高,LDH釋放量進一步減少,細胞凋亡率進一步降低,同時Caspase-3蛋白表達進一步減少(P0.05或P0.01)。以上指標相比,差異均有統(tǒng)計學意義。結(jié)論:梔子苷與美滿霉素聯(lián)合應用較單一應用能更有效拮抗PC12細胞凋亡,其機制可能與抑制Caspase-3蛋白表達有關(guān)。
[Abstract]:Aim: to observe the effect of geniposide combined with minocycline on apoptosis of PC12 cells induced by H_2O_2 and its mechanism. Methods: PC12 cells at logarithmic growth stage were randomly divided into control group, model group, geniposide group, minocycline group and combined application of geniposide and minocycline. The apoptosis model of PC12 cells induced by 300 渭 mol/L H_2O_2 for 6 h was established. The survival rate of PC12 cells was measured by MTT method, and the activity of lactate dehydrogenase (LDH) was measured by colorimetric method. Apoptosis was observed by inverted phase contrast microscope and fluorescence nuclear staining of Hoechest 33258. The apoptosis rate was detected by flow cytometry with Annexin V-FITC/PI double staining and the expression of Caspase-3 protein was detected by Western Blot. Results: compared with the model group, the survival rate of PC12 cells in geniposide group and minocycline group was decreased, the cell apoptosis rate was decreased, and the expression of Caspase-3 protein was decreased (P < 0.05), compared with those in geniposide group and minocycline group. In combination with geniposide / minocycline group, the survival rate of PC12 cells was further increased, the cell apoptosis rate was further decreased, and the expression of Caspase-3 protein was further decreased by P0.05 or P0.01. Compared with the above indexes, the differences were statistically significant. Conclusion: the combination of geniposide and minocycline can antagonize the apoptosis of PC12 cells more effectively than that of minocycline alone, and the mechanism may be related to the inhibition of Caspase-3 protein expression.
