抑制谷氨酰胺合成酶對(duì)小鼠紋狀體氨基酸類和單胺類遞質(zhì)含量的影響

【摘要】:目的:探討抑制谷氨酰胺合成酶對(duì)小鼠紋狀體氨基酸類遞質(zhì)和單胺類遞質(zhì)穩(wěn)態(tài)的影響。方法:3月齡C57BL/6J小鼠,腹腔給予谷氨酰胺合成酶抑制劑L-蛋氨酸磺酸鹽0.1 mL/10 g,隔日給藥,持續(xù)2周。Western blot方法檢測(cè)紋狀體谷氨酰胺合成酶以及谷氨酸轉(zhuǎn)運(yùn)體蛋白表達(dá)水平,高效液相色譜法檢測(cè)紋狀體谷氨酸、谷氨酰胺等氨基酸類遞質(zhì)和多巴胺、5-羥色胺等單胺類遞質(zhì)的含量。結(jié)果:同生理鹽水對(duì)照組相比,實(shí)驗(yàn)組小鼠紋狀體谷氨酸轉(zhuǎn)運(yùn)體1、谷氨酸天冬氨酸轉(zhuǎn)運(yùn)體表達(dá)上調(diào),谷氨酰胺合成酶表達(dá)下調(diào);谷氨酸、谷氨酰胺、天冬氨酸含量減少,γ-氨基丁酸、絲氨酸含量增加,而單胺類遞質(zhì)含量無(wú)明顯變化。結(jié)論:系統(tǒng)性給予谷氨酰胺合成酶抑制劑能影響腦內(nèi)氨基酸類遞質(zhì)代謝的穩(wěn)態(tài),導(dǎo)致興奮性神經(jīng)遞質(zhì)谷氨酸水平下降,抑制性神經(jīng)遞質(zhì)γ-氨基丁酸水平上升,但對(duì)單胺類遞質(zhì)含量沒(méi)有影響。
[Abstract]:Aim: to investigate the effects of inhibition of glutamine synthase on the homeostasis of amino acid transmitters and monoamine transmitters in the striatum of mice. Methods C57BL/6J mice of 3 months old were administered intraperitoneally with L-methionine sulfonate 0.1 mL/10 / g for 2 weeks. The expression of glutamine synthase and glutamate transporter protein in striatum was detected by Western blot. The contents of amino acid transmitters such as glutamate, glutamine and dopamine 5-hydroxytryptamine in striatum were determined by HPLC. Results: compared with the normal saline control group, the expression of glutamate transporter 1 and aspartate transporter in the striatum of the experimental group were up-regulated, the expression of glutamine synthase was down-regulated, the expression of glutamate, glutamine, glutamine, Aspartic acid content decreased, 緯-aminobutyric acid, serine content increased, but monoamine transmitter content did not change significantly. Conclusion: systemic administration of glutamine synthase inhibitors can affect the homeostasis of amino acid transmitters in the brain, resulting in a decrease in the level of excitatory neurotransmitter glutamate and an increase in the level of inhibitory neurotransmitter 緯 -aminobutyric acid. But there was no effect on the content of monoamine transmitters.
