瑞替加濱對離體大鼠十二指腸平滑肌收縮功能的影響及其機制

【摘要】:目的探討瑞替加濱對離體大鼠十二指腸平滑肌收縮功能的影響及其機制。方法取大鼠十二指腸腸管剪成長度約1.5 cm的腸段,采用BL-420F生物機能實驗系統(tǒng)持續(xù)記錄張力曲線。十二指腸腸管張力曲線穩(wěn)定后行以下試驗:(1)依次予3.125、6.25、12.5、25、50、100、150、200μmol/L的瑞替加濱溶液干預,每個濃度作用時間均為15 min,中間用Kreb's溶液沖洗20 min。測量瑞替加濱干預前及各濃度干預后十二指腸腸管平滑肌張力曲線的平均振幅。(2)加入10μmol/L XE-991作用15 min,再加入20μmol/L瑞替加濱作用15 min。計算瑞替加濱加藥前后離體十二指腸腸管平滑肌張力曲線的平均振幅變化百分率。(3)加入20μmol/L瑞替加濱作用15 min,再加入10μmol/L XE-991作用15 min。比較XE-991加藥前后離體十二指腸腸管平滑肌張力曲線的平均振幅變化百分率。結果 (1)瑞替加濱作用下離體十二指腸腸管平滑肌張力曲線的平均振幅(基線值)變化百分率為100%。各濃度瑞替加濱作用下張力曲線的平均振幅變化百分率均低于基線值,P均0.05。在6.25~200μmol/L范圍內(nèi),隨著瑞替加濱濃度增加提高,平均振幅變化百分率逐漸下降,P均0.05。(2)瑞替加濱加藥前后XE-991孵育的離體十二指腸腸管平滑肌張力曲線的平均振幅變化百分率無統(tǒng)計學差異,P0.05。(3)XE-991加藥后瑞替濱孵育的離體十二指腸腸管平滑肌張力曲線的平均振幅高于加藥前,P0.05。結論瑞替加濱在6.25~200μmol/L范圍內(nèi)可呈劑量依賴性抑制大鼠離體十二指腸平滑肌收縮;其機制可能為激活鉀離子通道。
[Abstract]:Objective to investigate the effect and mechanism of retegabine on contractile function of isolated rat duodenal smooth muscle. Methods the tension curves were recorded continuously by BL-420F biological function experiment system in the intestinal segment of rat duodenum which had a growing degree of 1.5 cm. After the tension curve of duodenum was stabilized, the following experiment: 1: 1) was treated with retegabine solution of 3.125 ~ 6.25 ~ 12.5 ~ (12.5) ~ 2550 ~ 100 ~ 150200 渭 mol/L. The time of treatment for each concentration was 15 minutes, and the Kreb's solution was used to wash the solution for 20 minutes. The mean amplitude of tension curve of duodenal smooth muscle was measured before and after the intervention of retegabine. The mean amplitude of tension curve of duodenal smooth muscle was measured. 10 渭 mol/L XE-991 was added for 15 minutes, and then 20 渭 mol/L retegabine was added for 15 minutes. The mean amplitude change percentage of tension curve of isolated duodenal smooth muscle was calculated before and after retegabine was added. (3) 20 渭 mol/L retegabine was added for 15 minutes, and then 10 渭 mol/L XE-991 was added for 15 minutes. To compare the average amplitude percentage of tension curve of isolated duodenal smooth muscle before and after administration of XE-991. Results 1) the percentage of mean amplitude (baseline value) of tension curve of isolated duodenal smooth muscle under retegabine was 100%. The percentage of mean amplitude change of tension curve under the action of retegabine was lower than that of baseline value (P < 0.05). In the range of 6.25 渭 mol/L, with the increase of retegabine concentration, The percentage of mean amplitude change decreased gradually (P < 0.05. 2) the average amplitude change percentage of tension curve of isolated duodenal smooth muscle incubated with XE-991 before and after retegabine administration had no statistical difference. The mean amplitude of smooth muscle tension curve of somatoduodenum was higher than that of P0.05 before administration. Conclusion Retigabine can inhibit the contraction of isolated rat duodenal smooth muscle in a dose-dependent manner in the range of 6.25 渭 mol/L, and the mechanism may be the activation of potassium channel.
