口服富氫水對小鼠脂肪肝缺血再灌注損傷的保護(hù)作用

【摘要】:目的探討富氫水對脂肪肝缺血再灌注損傷小鼠模型的保護(hù)作用。方法將30只C57BL/6小鼠隨機(jī)分為3組。假手術(shù)組:喂蛋氨酸膽堿缺乏高脂飲食非酒精性脂肪肝造模;對照組:肝臟缺血再灌注前灌服蒸餾水;富氫水缺血再灌注組:肝臟缺血再灌注前灌服富氫水,再灌注6 h后處死小鼠,檢測血清中AST和ALT含量,觀察肝臟組織形態(tài)學(xué)變化,分析肝臟組織損傷程度、巨噬細(xì)胞浸潤、炎性因子TNF-α、IL-6、OPN mRNA表達(dá)水平和肝臟組織內(nèi)活化的Caspase-3蛋白表達(dá)水平。結(jié)果與對照組相比較,富氫水組血清ALT和AST水平較低(P0.05),巨噬細(xì)胞浸潤和凋亡細(xì)胞明顯減少,炎性因子mRNA的表達(dá)明顯降低。Caspase-3表達(dá)明顯降低。結(jié)論富氫水能夠減輕脂肪肝小鼠肝臟缺血再灌注損傷,其機(jī)制與富氫水抑制氧化應(yīng)激反應(yīng)和炎癥反應(yīng),進(jìn)而抑制Caspase-3蛋白表達(dá)有關(guān)。
[Abstract]:Objective to investigate the protective effect of hydrogen rich water on fatty liver ischemia reperfusion injury in mice. Methods 30 C57BL/6 mice were randomly divided into 3 groups. Sham-operation group: model of non-alcoholic fatty liver induced by methionine choline deficiency diet; control group: distilled water before hepatic ischemia-reperfusion; Hydrogen-rich water ischemia-reperfusion group: before liver ischemia-reperfusion, After reperfusion for 6 h, the mice were killed, the contents of AST and ALT in serum were measured, the changes of liver histomorphology were observed, the degree of liver injury and the infiltration of macrophages were analyzed. The expression level of inflammatory factor TNF- 偽 and IL-6 mRNA and the expression of activated Caspase-3 protein in liver tissue. Results compared with the control group, the levels of serum ALT and AST were lower, macrophage infiltration and apoptotic cells were significantly decreased, and the expression of inflammatory factor mRNA was significantly decreased in the hydrogen-rich water group than that in the control group. Conclusion Hydrogen-rich water can attenuate hepatic ischemia-reperfusion injury in fatty liver mice, and its mechanism is related to the inhibition of oxidative stress and inflammation by hydrogen-rich water, and thus the inhibition of Caspase-3 protein expression.
