內(nèi)毒素耐受對細胞因子的影響

【摘要】:目的進一步明確內(nèi)毒素耐受時炎性相關(guān)細胞因子的變化。方法昆明鼠分組處置。建立內(nèi)毒素(LPS)耐受鼠模型。ELISA方法測定血清腫瘤壞死因子α(TNF-α),白細胞介素10(IL-10)和白細胞介素17(IL-17)濃度。結(jié)果 LPS對照組血清TNF-α、IL-10和IL-17濃度分別為(2 261.49±1 090.28)pg/m L,(7 780.42±2 320.04)pg/m L,(27.28±13.76)pg/m L,較健康對照組顯著增高。而內(nèi)毒素耐受組相應(yīng)細胞因子血清濃度分別為(1 174.19±450.91)pg/m L,(1 293.26±520.85)pg/m L,(22.26±11.44)pg/m L。地塞米松干預(yù)進一步降低了TNF-α和IL-17的水平,但對IL-10水平無顯著影響。結(jié)論內(nèi)毒素耐受狀態(tài)與內(nèi)毒素炎癥狀態(tài)比較,TNF-α和IL-10水平顯著降低,IL-17水平無顯著變化。應(yīng)用地塞米松可進一步降低內(nèi)毒素耐受狀態(tài)下的TNF-α和IL-17水平。
[Abstract]:Objective to investigate the changes of inflammatory cytokines during endotoxin tolerance. Methods Kunming rats were divided into groups. A rat model of endotoxin tolerance was established. The serum levels of TNF- 偽, IL-10 and IL-17 were determined by Elisa. Results the serum levels of TNF- 偽 IL-10 and IL-17 in LPS group were 2 261.49 鹵1 090.28)pg/m / L, 7 780.42 鹵2 320.04)pg/m / L and 27.28 鹵13.76)pg/m / L, respectively, which were significantly higher than those in healthy control group. In endotoxin tolerance group, the serum concentration of cytokines was 1 174.19 鹵450.91)pg/m / L, 1 293.26 鹵520.85)pg/m / L, 22.26 鹵11.44)pg/m / L, respectively. Dexamethasone further reduced the levels of TNF- 偽 and IL-17, but had no significant effect on IL-10 levels. Conclusion the levels of TNF- 偽 and IL-10 in endotoxin tolerance state and endotoxin inflammation state were significantly lower than that in endotoxin inflammatory state. Dexamethasone could further reduce the levels of TNF- 偽 and IL-17 in endotoxin tolerant state.
