基于生物質(zhì)譜的蛋白質(zhì)定量技術(shù)及其在藥代動力學(xué)研究中的應(yīng)用

【摘要】:蛋白質(zhì)定量技術(shù)是指對細(xì)胞、組織以至整體生物體內(nèi)的蛋白質(zhì)表達(dá)水平進(jìn)行全譜定量分析的手段,其在生物過程機(jī)制的探索、生物標(biāo)志物和藥物作用靶標(biāo)的發(fā)現(xiàn)及驗(yàn)證等過程中發(fā)揮著重要作用。新興生物質(zhì)譜技術(shù)和數(shù)據(jù)采集方法的發(fā)展為蛋白質(zhì)定量提供了先進(jìn)的技術(shù)平臺,促使了蛋白質(zhì)研究逐漸從簡單定性轉(zhuǎn)變?yōu)榫_定量。本文綜述了基于生物質(zhì)譜的蛋白分子定量策略及方法,評述了不同方法的特點(diǎn)和適用范圍,并闡述了其在藥物代謝動力學(xué)研究領(lǐng)域,如藥物轉(zhuǎn)運(yùn)體、藥物代謝酶的定量和多肽及蛋白類藥物的藥代動力學(xué)研究等方面的應(yīng)用,為傳統(tǒng)的藥代動力學(xué)研究提供了新的思路與方法。
[Abstract]:Protein quantitative technique refers to the means of full spectrum quantitative analysis of protein expression level in cells, tissues and even the whole organism, and the exploration of its mechanism in biological process. The discovery and verification of biomarkers and drug targets play an important role. The development of new biological mass spectrometry and data acquisition methods provides an advanced technology platform for protein quantification, which promotes protein research to change from simple qualitative analysis to accurate quantification. In this paper, the strategies and methods of protein molecular quantification based on bio-mass spectrometry are reviewed. The characteristics and application of different methods are reviewed, and their applications in the field of pharmacokinetics, such as drug transporters, are reviewed. The application of quantification of drug metabolizing enzymes and pharmacokinetics of peptides and proteins provides a new way of thinking and method for traditional pharmacokinetic research.
