microRNA對(duì)藥物體內(nèi)過(guò)程調(diào)控的研究進(jìn)展

【摘要】:藥物的體內(nèi)過(guò)程需經(jīng)一系列的生物轉(zhuǎn)化和轉(zhuǎn)運(yùn)途徑,依賴(lài)于藥物代謝酶和轉(zhuǎn)運(yùn)體的參與。而個(gè)體對(duì)同一藥物的代謝、轉(zhuǎn)運(yùn)能力存在差異,這一差異不能完全用藥物基因組學(xué)解釋。microRNA作為表觀(guān)遺傳修飾的一個(gè)重要方面,是對(duì)傳統(tǒng)遺傳學(xué)的強(qiáng)有力補(bǔ)充。人體內(nèi)多種藥物代謝酶和轉(zhuǎn)運(yùn)體均受到不同的microRNA調(diào)控,同一microRNA又可同時(shí)調(diào)控不同的代謝酶或(和)轉(zhuǎn)運(yùn)體,二者均提示microRNA極有可能實(shí)現(xiàn)較為廣泛的宏觀(guān)調(diào)控。該文分別從microRNA對(duì)藥物代謝酶的調(diào)控、對(duì)藥物轉(zhuǎn)運(yùn)體的調(diào)控以及同時(shí)調(diào)控代謝酶及轉(zhuǎn)運(yùn)體的microRNA三個(gè)方向綜合分析,為研究藥物個(gè)體差異提出一個(gè)極好的切入點(diǎn),并為合理用藥和個(gè)體化醫(yī)療提供理論基礎(chǔ)。
[Abstract]:A series of biotransformation and translocation pathways are required for the in vivo process of drugs, which depend on the involvement of drug metabolizing enzymes and transporters. However, individual metabolism and transport ability of the same drug are different. This difference can not be fully explained by pharmacogenomics. MicroRNA is an important aspect of epigenetic modification, and it is a powerful supplement to traditional genetics. Many drug metabolic enzymes and transporters in human body are regulated by different microRNA, and the same microRNA can regulate different metabolic enzymes or / and transporters at the same time. Both of them suggest that microRNA is likely to achieve a wide range of macro-regulation. In this paper, we analyze the regulation of drug metabolic enzymes by microRNA, the regulation of drug transporters and the regulation of metabolic enzymes and transporters by microRNA, respectively, which provides an excellent starting point for the study of drug individual differences. It also provides theoretical basis for rational drug use and individualized medical treatment.
