生長激素釋放肽對大鼠脊髓缺血再灌注損傷的保護(hù)作用

【摘要】:目的探討生長激素釋放肽對大鼠脊髓缺血再灌注損傷的保護(hù)作用。方法將60只SD大鼠分為假手術(shù)組、損傷組、藥物組,每組20只。損傷組和藥物組以夾閉腹主動脈法制作大鼠缺血再灌注模型,假手術(shù)組不夾閉腹主動脈。術(shù)后,假手術(shù)組和損傷組大鼠腹腔注射生理鹽水,藥物組大鼠腹腔注射生長激素釋放肽。分別于術(shù)后12、24、48和72 h取材,RT-PCR檢測脊髓損傷區(qū)組織生長激素促分泌素受體-1α(GHSR-1α)mRNA的表達(dá),酶聯(lián)免疫吸附法測定血清腫瘤壞死因子-α(TNF-α)含量及髓過氧化物酶(MPO)活性,TUNEL法檢測脊髓損傷區(qū)細(xì)胞凋亡情況,Western blot檢測脊髓損傷區(qū)組織Bax、Bcl-2蛋白的表達(dá)。結(jié)果損傷組脊髓組織GHSR-1αmRNA表達(dá)量較假手術(shù)組明顯下調(diào)(P0.05),藥物組脊髓組織GHSR-1αmRNA表達(dá)量較損傷組明顯上調(diào)(P0.05)。術(shù)后各時點(diǎn),損傷組血清TNF-α含量及脊髓組織MPO活性均較假手術(shù)組明顯增加(P0.05),而藥物組血清TNF-α含量及脊髓組織MPO活性均較損傷組明顯減小(P0.05)。術(shù)后24 h,藥物組脊髓組織神經(jīng)細(xì)胞凋亡率明顯低于損傷組(P0.05)。術(shù)后各時點(diǎn),藥物組與損傷組脊髓組織Bax蛋白表達(dá)量差異均無統(tǒng)計學(xué)意義(P0.05),而術(shù)后24、48 h時藥物組脊髓組織Bcl-2蛋白表達(dá)量均較損傷組上調(diào)(P0.01)。結(jié)論生長激素釋放肽對大鼠脊髓缺血再灌注損傷具有保護(hù)作用,該作用與減少神經(jīng)細(xì)胞炎癥反應(yīng)及凋亡等有關(guān)。
[Abstract]:Objective to investigate the protective effect of growth hormone releasing peptide (GH) on spinal cord ischemia reperfusion injury in rats. Methods Sixty SD rats were divided into sham-operation group, injury group and drug group with 20 rats in each group. The ischemia reperfusion model of rats was made by clamping abdominal aorta in injury group and drug group, and no clamping of abdominal aorta was observed in sham operation group. After operation, rats in sham operation group and injury group were intraperitoneally injected with normal saline, and rats in drug group were intraperitoneally injected with growth hormone releasing peptide. RT-PCR was used to detect the expression of growth hormone secretin receptor 1 偽 (GHSR-1 偽) mRNA in the injured area of spinal cord. The levels of serum tumor necrosis factor- 偽 (TNF- 偽) and the activity of myeloperoxidase (MPO) were determined by enzyme-linked immunosorbent assay (Elisa) and Tunel method was used to detect apoptosis in spinal cord injury area. Western blot was used to detect the expression of Baxfen Bcl-2 protein in spinal cord injury area. Results the expression of GHSR-1 偽 mRNA in spinal cord in the injured group was significantly lower than that in the sham operation group, and the expression of GHSR-1 偽 mRNA in the spinal cord in the drug group was significantly higher than that in the injured group. At all time points after operation, the content of TNF- 偽 in serum and the activity of MPO in spinal cord in the injured group were significantly higher than those in the sham operation group, while the content of TNF- 偽 in the serum and the MPO activity in the spinal cord in the drug group were significantly lower than those in the injured group. 24 hours after operation, the apoptotic rate of nerve cells in the spinal cord in the drug group was significantly lower than that in the injured group (P 0.05). At each time point after operation, there was no significant difference in the expression of Bax protein between the drug group and the injured group. However, at 24 h after operation, the expression of Bcl-2 protein in the drug group was significantly higher than that in the injured group (P 0.01). Conclusion growth hormone releasing peptide has protective effect on spinal cord ischemia-reperfusion injury in rats, which is related to the reduction of inflammatory reaction and apoptosis of nerve cells.
