【摘要】：目的研究索拉非尼對(duì)宮頸癌He La細(xì)胞增殖的抑制作用,并對(duì)其抑癌機(jī)制進(jìn)行初步探討。方法用終濃度為0.01,0.1,1.0,2.5,5.0 mg·L~(-1)的索拉非尼干預(yù)宮頸癌He La細(xì)胞(實(shí)驗(yàn)組),無(wú)任何藥物處理的宮頸癌He La細(xì)胞為對(duì)照組,培養(yǎng)1,2,3 d后,用溴化四唑藍(lán)比色(MTT)法檢測(cè)各組宮頸癌He La細(xì)胞增殖情況,用細(xì)胞免疫熒光法檢測(cè)及酶聯(lián)免疫吸附實(shí)驗(yàn)(ELISA)法檢測(cè)各組血管內(nèi)皮生長(zhǎng)因子-C(VEGF-C)和磷酸化細(xì)胞外信號(hào)調(diào)節(jié)激酶(p-ERK)蛋白表達(dá)情況。結(jié)果培養(yǎng)3 d后,對(duì)照組、0.01,0.1,1.0,2.5,5.0 mg·L~(-1)實(shí)驗(yàn)組的細(xì)胞增殖抑制率分別為(5.15±1.67)%,(9.16±1.58)%,(23.07±1.86)%,(30.86±2.67)%,(55.94±3.92)%,(68.75±4.48)%。實(shí)驗(yàn)組與對(duì)照組比較,差異有統(tǒng)計(jì)學(xué)意義(P0.01)。實(shí)驗(yàn)組細(xì)胞培養(yǎng)3 d的增殖抑制率與培養(yǎng)1d比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。細(xì)胞免疫熒光檢測(cè)結(jié)果顯示,對(duì)照組中綠色熒光明顯增強(qiáng),經(jīng)不同濃度索拉非尼處理后,綠色熒光明顯減弱。對(duì)照組、0.01,0.1,1.0 mg·L~(-1)實(shí)驗(yàn)組的VEGF-C分別為(1326.54±139.57),(1305.64±119.82),(1235.57±102.34),(1083.33±89.61)pg·m L~(-1);對(duì)照組、0.01,0.1,1.0 mg·L~(-1)實(shí)驗(yàn)組的p-ERK分別為(2.34±0.28),(2.21±0.22),(1.56±0.21),(0.96±0.13)μg·m L~(-1)。實(shí)驗(yàn)組的VEGF-C和p-ERK蛋白表達(dá)量明顯低于對(duì)照組(P0.05),且VEGF-C和p-ERK的表達(dá)隨藥物濃度的增加而逐漸降低,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論索拉非尼可抑制宮頸癌He La細(xì)胞增殖,且抑制率與其作用時(shí)間、濃度呈正比,其抑癌機(jī)制可能與其能夠抑制宮頸癌He La細(xì)胞VEGF-C和p-ERK蛋白表達(dá)相關(guān)。
[Abstract]:Objective to study the inhibitory effect of Solafenib on the proliferation of cervical cancer cell line he La and its mechanism. Methods Sorafenil (final concentration was 0.01g / ml) was used to interfere with cervical cancer He-La cells (experimental group, control group, without any drug treatment), and cultured for 1 day, 2 days later, after 3 days of culture, helafenil was used to treat cervical cancer He-La cells, the control group was treated with helafenil as the control group, and the He-La cells were cultured for 3 days. The proliferation of helium La cells in each group was detected by MTT assay. The expression of vascular endothelial growth factor (VEGF-C) and phosphorylated extracellular signal-regulated kinase (pERK) protein were detected by cell immunofluorescence assay and enzyme linked immunosorbent assay (Elisa). Results after 3 days of culture, the cell proliferation inhibition rate of the control group was 0.01 鹵1.50 mg / L, and the inhibitory rate was 5.15 鹵1.67 and 23.07 鹵1.866.86 鹵3.922 鹵3.922 鹵4.48. The inhibition rate of cell proliferation in the experimental group was 55.94 鹵3.922 and 68.75 鹵4.48, respectively. The difference between the experimental group and the control group was statistically significant (P 0.01). The proliferation inhibition rate of the experimental group was significantly higher than that of the 1 day culture group (P 0.05). The results of cell immunofluorescence assay showed that the green fluorescence in the control group was significantly increased, and the green fluorescence was significantly decreased after different concentrations of Solafenib. The VEGF-C of the control group was 1326.54 鹵139.57 鹵1305.64 鹵119.82 鹵1235.57 鹵1083.33 鹵1083.33 鹵1083.33 鹵89.61)pg / L, respectively, and the p-ERK of the control group was 2.34 鹵0.28 mg / L, respectively. The p-ERK of the experimental group was 2.34 鹵0.28 (1.56 鹵0.21 鹵0.96 鹵0.13) 渭 g / mL. The expression of VEGF-C and p-ERK protein in the experimental group was significantly lower than that in the control group (P 0.05), and the expression of VEGF-C and p-ERK decreased gradually with the increase of drug concentration. The difference was statistically significant (P 0.05). Conclusion Solafenil can inhibit the proliferation of He-La cells, and the inhibition rate is directly proportional to the time and concentration of the inhibition. The mechanism of the inhibition may be related to the inhibition of the expression of VEGF-C and p-ERK proteins in He-La cells.
