厄貝沙坦對(duì)大鼠梗死心肌組織中HGF mRNA和蛋白水平的影響

【摘要】:目的:通過觀察厄貝沙坦對(duì)大鼠心肌梗死模型中缺血心肌肝細(xì)胞生長(zhǎng)因子(HGF)mRNA和蛋白水平的影響,探討厄貝沙坦減輕心肌纖維化的可能機(jī)制。方法:采用雄性Wistar大鼠建立心肌梗死模型,術(shù)后24 h將存活大鼠隨機(jī)分為模型組和厄貝沙坦組,另設(shè)假手術(shù)組,每組9只大鼠。厄貝沙坦組予以厄貝沙坦50 mg·kg-1·d-1灌胃,模型組及假手術(shù)組給予等體積生理鹽水灌胃,每天1次。4周后心臟取材,測(cè)量各組大鼠體重及左室重量,HE染色法觀察缺血心肌病理改變,RT-q PCR和Western blot法檢測(cè)各組大鼠心肌組織中HGF mRNA及蛋白水平的表達(dá)。結(jié)果:HE染色可見假手術(shù)組心肌細(xì)胞排列整齊,模型組及厄貝沙坦組心肌結(jié)構(gòu)紊亂,藥物組病理改變較模型組減輕;4周后各組大鼠體重?zé)o顯著差異,各組間左心室重量差異有統(tǒng)計(jì)學(xué)意義(P0.01),厄貝沙坦組及模型組左室重量高于假手術(shù)組(P0.05),同時(shí)厄貝沙坦組低于模型組(P0.05);各組大鼠心肌組織中均檢測(cè)到HGF的mRNA及蛋白水平,厄貝沙坦組及模型組HGF的mRNA與蛋白含量均高于假手術(shù)組(P0.05),厄貝沙坦組HGF的mRNA與蛋白水平低于模型組(P0.05)。結(jié)論:心肌梗死模型大鼠應(yīng)用厄貝沙坦治療4周時(shí)左室重量明顯減輕,心肌組織病理變化得到改善,心肌組織中HGF的mRNA與蛋白水平降低。
[Abstract]:Aim: to investigate the effect of irbesartan on the mRNA and protein levels of hepatocyte growth factor (HGFN) in ischemic myocardium of rats with myocardial infarction, and to explore the possible mechanism of irbesartan in alleviating myocardial fibrosis. Methods: male Wistar rats were used to establish myocardial infarction model. The surviving rats were randomly divided into model group and irbesartan group 24 hours after operation. Irbesartan group was given irbesartan 50 mg kg-1 d-1 intragastrically, model group and sham operation group were given the same volume of normal saline. The pathological changes of ischemic myocardium were observed by HE staining and RT-Q PCR and Western blot were used to detect the expression of HGF mRNA and protein in myocardial tissue of rats in each group. Results the myocardial cells were arranged neatly in the sham-operation group, and the myocardial structure in the model group and irbesartan group were disordered. The pathological changes in the drug group were not significantly different from those in the model group after 4 weeks. The left ventricular weight of irbesartan group and model group was higher than that of sham-operated group (P 0.05), and irbesartan group was lower than model group (P 0.05). The mRNA and protein levels of HGF were detected in myocardium of all groups. The contents of mRNA and protein in HGF of irbesartan group and model group were higher than those of sham operation group (P 0.05), and the mRNA and protein levels of HGF in irbesartan group were lower than those in model group (P 0.05). Conclusion: after treatment with irbesartan for 4 weeks, left ventricular weight was significantly reduced, myocardial histopathological changes were improved, and the mRNA and protein levels of HGF in myocardial tissue were decreased in myocardial infarction model rats.
