微波加熱順鉑還原鐵粉混懸液對豬肝細(xì)胞凋亡的影響

【摘要】:【目的】 探討微波加熱順鉑還原鐵粉混懸液誘導(dǎo)豬肝細(xì)胞凋亡及其與TNF-α、Bcl-2和Bax蛋白表達的關(guān)系。 【方法】 將豬隨機地分成對照組和微波治療組。微波治療組用穿刺針將順鉑還原鐵粉混懸液注入消融治療后的豬肝形成的空洞內(nèi),用醫(yī)用微波治療儀局部加熱,對照組則不予處理。采用原位末端轉(zhuǎn)移酶標(biāo)記(TUNEL)技術(shù)分析檢測細(xì)胞的凋亡情況,免疫組化法檢測細(xì)胞凋亡相關(guān)基因TNF-α、Bcl-2和Bax蛋白的表達水平。 【結(jié)果】 1、微波治療組和對照組豬肝內(nèi)均可見凋亡細(xì)胞,微波治療組中平均凋亡指數(shù)(%)為7.55±1.74,對照組中平均凋亡指數(shù)(%)為5.33±1.45;兩者相比較,微波治療組細(xì)胞平均凋亡指數(shù)(%)明顯高于對照組,差異有統(tǒng)計學(xué)意義(P0.01)。 2、微波治療組中TNF-α蛋白平均陽性表達率(%)為11.70±3.02,對照組中TNF-α蛋白平均陽性表達率(%)為8.20±1.43;兩者相比較,微波治療組TNF-α蛋白陽性表達率明顯高于對照組,,差異有統(tǒng)計學(xué)意義(P0.01)。 3、微波治療組中Bcl-2蛋白平均陽性表達率(%)為6.61±1.74,對照組中Bcl-2蛋白平均陽性表達率(%)為9.81±3.01;兩者相比較,微波治療組Bcl-2蛋白平均陽性表達率較對照組明顯降低,差異有統(tǒng)計學(xué)意義(P0.05)。 4、微波治療組中Bax蛋白平均陽性表達率(%)為13.37±3.40,對照組中Bax蛋白平均陽性表達率(%)為9.81±2.80,兩者相比較,微波治療組Bax蛋白平均陽性表達率較對照組明顯升高,差異有統(tǒng)計學(xué)意義(P0.05)。 5、微波治療組中Bax/Bcl-2平均比值為5.34±2.55,對照組中Bax/Bcl-2平均比值為3.29±1.59;兩者相比較,微波治療組Bax/Bcl-2平均比值較對照組明顯升高,差異有統(tǒng)計學(xué)意義(P0.05)。 【結(jié)論】 1、微波加熱順鉑還原鐵粉混懸液能有效地誘導(dǎo)豬肝細(xì)胞凋亡。 2、微波加熱順鉑還原鐵粉混懸液能有效地誘導(dǎo)豬肝細(xì)胞凋亡的機制可能是通過以下幾條途徑來實現(xiàn)的: 1)增加TNF-α蛋白的表達; 2)抑制Bcl-2蛋白的表達; 3)增加Bax蛋白的表達; 4)提高Bax/Bcl-2的比值。
[Abstract]:[purpose] To investigate the relationship between the apoptosis of porcine hepatocytes induced by Cisplatin reduced iron powder suspension and the expression of TNF- 偽 Bcl-2 and Bax protein. [methods] Pigs were randomly divided into control group and microwave treatment group. In the microwave treatment group, Cisplatin reduced iron powder suspension was injected into the cavity of the pig liver after ablation with puncture needle, and the local heating was used with the medical microwave therapy instrument, while the control group was not treated. Apoptosis was detected by in situ terminal transferase labeling (Tunel) technique and the expression of apoptosis related gene TNF- 偽 and Bax protein was detected by immunohistochemistry. [results] 1. Apoptotic cells were found in the liver of pigs in microwave treatment group and control group, the average apoptosis index in microwave treatment group was 7.55 鹵1.74, and the average apoptosis index in control group was 5.33 鹵1.45.The mean apoptotic index in microwave treatment group was significantly higher than that in control group. The difference was statistically significant (P 0.01). (2) the average positive expression rate of TNF- 偽 protein in microwave treatment group was 11.70 鹵3.02, and that in control group was 8.20 鹵1.43.The positive rate of TNF- 偽 protein expression in microwave treatment group was significantly higher than that in control group (P 0.01). 3. The average positive expression rate of Bcl-2 protein was 6.61 鹵1.74 in microwave treatment group and 9.81 鹵3.01 in control group. Compared with control group, the average positive rate of Bcl-2 protein in microwave treatment group was significantly lower than that in control group (P 0.05). 4. The average positive expression rate of Bax protein was 13.37 鹵3.40 in microwave group and 9.81 鹵2.80 in control group. Compared with the control group, the average positive rate of Bax protein in microwave treatment group was significantly higher than that in control group (P 0.05). 5, the average ratio of Bax/Bcl-2 in microwave treatment group was 5.34 鹵2.55, and that in control group was 3.29 鹵1.59.The average ratio of Bax/Bcl-2 in microwave treatment group was significantly higher than that in control group (P 0.05). [conclusion] 1. Microwave heating of Cisplatin reduced iron powder suspension could effectively induce the apoptosis of porcine hepatocytes. 2. Microwave heating of Cisplatin reduced iron powder suspension can effectively induce the apoptosis of porcine hepatocytes. 1) increase the expression of TNF- 偽 protein; 2) inhibiting the expression of Bcl-2 protein; 3) increase the expression of Bax protein; 4) increasing the ratio of Bax/Bcl-2.
