氧化釹經(jīng)鼻滴注對(duì)小鼠神經(jīng)系統(tǒng)的影響

【摘要】:研究氧化釹經(jīng)鼻滴注對(duì)小鼠神經(jīng)系統(tǒng)的影響,進(jìn)一步考察氧化釹的神經(jīng)毒性是否存在劑量和尺寸效應(yīng)。選取健康的ICR雌鼠48只,隨機(jī)分為對(duì)照組、低劑量納米(L-nm)氧化釹組(1μg/mL)、高劑量納米(H-nm)氧化釹組(40μg/mL)和低劑量微米(L-μm)氧化釹組(1μg/mL)。經(jīng)鼻染毒,共10d,隔天1次;染毒第2次之后進(jìn)行水迷宮試驗(yàn),定位航行試驗(yàn)7d,1d一個(gè)單元,同時(shí)在第7天進(jìn)行空間探索試驗(yàn);取血,進(jìn)行生化指標(biāo)檢測(cè);取腦置于100g/L甲醛溶液固定,常規(guī)脫水,石蠟包埋、切片,蘇木素-伊紅(HE)染色。劑量效應(yīng)顯示,不同濃度納米氧化釹對(duì)小鼠經(jīng)鼻滴注,高納米組與低納米組相比,目標(biāo)象限游程占總游程百分比較高(P0.05),但在此尺寸條件下劑量效應(yīng)不明顯。尺寸效應(yīng)顯示,相同濃度納米氧化釹對(duì)小鼠經(jīng)鼻滴注,L-μm組海馬顆粒細(xì)胞排列稀疏,細(xì)胞脫落;L-nm組海馬齒狀回顆粒細(xì)胞排列較整齊,可見部分細(xì)胞核固縮在此試驗(yàn)濃度條件下尺寸效應(yīng)不明顯。血液生物標(biāo)志物檢查試驗(yàn)顯示,氧化釹經(jīng)鼻滴注會(huì)對(duì)小鼠肝功能、腎功能產(chǎn)生一定的損傷。不同濃度納米氧化釹經(jīng)鼻滴注,對(duì)小鼠有輕微的刺激學(xué)習(xí)記憶的作用,但劑量效應(yīng)不明顯。低微米氧化釹對(duì)小鼠神經(jīng)系統(tǒng)有輕微的損傷,但與低納米氧化釹相比,差異不顯著,表明尺寸效應(yīng)不明顯。
[Abstract]:The effects of nasal drip of neodymium oxide on the nervous system of mice were studied and the dose and size effects of the neurotoxicity of neodymium oxide were investigated. 48 healthy female ICR rats were randomly divided into control group, low dose neodymium oxide group (1 渭 g / mL), high dose neodymium oxide group (40 渭 g / mL) and low dose neodymium oxide group (1 渭 g / mL). After the second exposure, water labyrinth test was carried out, a unit of navigation test was located for 7 days, and space exploration test was carried out on the 7th day, blood was taken and biochemical indexes were detected. The brain was fixed in 100g/L formaldehyde solution, dehydrated routinely, embedded in paraffin, sectioned and stained with hematoxylin-eosin. The dose effect showed that the percentage of target quadrant run in the total run was higher in the high nanometer group than in the low nanometer group, but the dose effect was not obvious under the condition of different concentrations of neodymium oxide. The size effect showed that the neodymium oxide at the same concentration was sparsely arranged in hippocampal granulosa cells in the L- 渭 m group, and that in the L-nm group, the granulosa cells in the dentate gyrus of the horse were arranged neatly. It can be seen that the size effect of partial nuclear pyknosis is not obvious at this experimental concentration. Blood biomarker test showed that neodymium oxide could damage liver and kidney function of mice. Different concentrations of nano-neodymium oxide by nasal drip can stimulate learning and memory in mice slightly, but the dose effect is not obvious. Low micron neodymium oxide had slight damage to the nervous system of mice, but the difference was not significant compared with that of low nanometer neodymium oxide, indicating that the size effect was not obvious.
