環(huán)磷酰胺對小鼠免疫抑制作用過程中代謝通路變化的代謝組學研究

【摘要】:目的采用代謝組學方法,研究環(huán)磷酰胺對小鼠免疫抑制作用過程中代謝通路的變化。方法采用流式細胞術了解免疫細胞變化情況,通過建立偏最小二乘法模型多結果進行判別。以偏最小二乘法模型中變量重要性投影(variable importance in projection,VIP)參數(shù)篩選潛在的生物標志物,通過統(tǒng)計分析確定差異性代謝物,借助代謝組學小分子化合物快速鑒定分析軟件系統(tǒng)對差異性代謝物進行進一步鑒定分析,通過搜庫及運算最終確定差異代謝物質(zhì)9種,通路富集結果分析差異代謝通路3條。結果環(huán)磷酰胺對正常機體的不飽和脂肪酸的生物合成、線粒體脂肪酸代謝、甘油磷脂代謝、甾類激素的生物合成、花生四烯酸代謝、脂肪酸代謝、嘧啶的生物合成等代謝通路均產(chǎn)生明顯影響。結論環(huán)磷酰胺在正常組與實驗組中影響最重要的代謝通路為花生四烯酸代謝、甘油磷脂代謝和嘧啶代謝。
[Abstract]:Objective to study the changes of metabolic pathway during immunosuppressive effect of cyclophosphamide in mice. Methods flow cytometry was used to investigate the changes of immune cells. The potential biomarkers were screened by the variable importance in the partial least square model, and the different metabolites were determined by statistical analysis. The differential metabolites were further identified and analyzed with the help of the rapid identification and analysis software system of metabolomics small molecular compounds. Nine different metabolites were identified by searching database and operation, and three different metabolic pathways were analyzed by the enrichment results of pathway. Results the biosynthesis of unsaturated fatty acids, mitochondrial fatty acid metabolism, glycerol phospholipid metabolism, steroid hormone biosynthesis, arachidonic acid metabolism and fatty acid metabolism were observed. Pyrimidine biosynthesis and other metabolic pathways have a significant effect. Conclusion the most important metabolic pathway of cyclophosphamide in normal group and experimental group is arachidonic acid metabolism, glycerol phospholipid metabolism and pyrimidine metabolism.
