【摘要】：目的建立測(cè)定人血漿中拉莫三嗪、奧卡西平及其代謝物10-羥基卡馬西平濃度的高效液相色譜方法。方法以非那西汀為內(nèi)標(biāo),血漿樣品用乙醚萃取,用C18柱分析,流動(dòng)相為乙腈-0.05 mol·L-1磷酸二氫鈉水溶液(25∶75,p H 4.5),流速為1.0 m L·min-1,柱溫為25℃,檢測(cè)波長(zhǎng)為220 nm。同時(shí)考察該方法的專屬性、標(biāo)準(zhǔn)曲線和定量下限、精密度與回收率、穩(wěn)定性。結(jié)果拉莫三嗪、10-羥基卡馬西平、奧卡西平分別在0.1~20.0μg·m L-1(y=20.11×10-2c+1.31×10-2,r=0.999 0),0.5~50.0μg·m L-1(y=9.77×10-2c+0.35×10-2,r=0.999 8),0.25~25.0μg·m L-1(y=131.69×10-2c-0.59×10-2,r=0.999 1)線性關(guān)系良好。拉莫三嗪、10-羥基卡馬西平、奧卡西平的方法回收率均在88.65%~100.38%,日內(nèi)、日間精密度均15%。結(jié)論該方法操作簡(jiǎn)便、準(zhǔn)確、靈敏度高,可用于拉莫三嗪、奧卡西平及其代謝物10-羥基卡馬西平血藥濃度監(jiān)測(cè)。
[Abstract]:Objective to establish a high performance liquid chromatography (HPLC) method for the determination of lamotriazine oxcarbazepine and its metabolite 10-hydroxycarbamazepine in human plasma.
