新型可控性栓塞劑MNPs-tTF的構(gòu)建

【摘要】:目的:構(gòu)建新型的具有可控性且能誘發(fā)動(dòng)物血栓的栓塞劑——磁靶向凝血蛋白,從而為動(dòng)物血栓模型的建立提供新工具。方法:使用化學(xué)共沉淀法合成磁流體(magnetic nanoparticles,MNPs),通過透析對其純化后用超導(dǎo)量子干涉磁強(qiáng)針(SQUID)測定其磁性能;利用基因工程方法表達(dá)截短組織因子(truncated tissue factor,tTF)(又稱為凝血蛋白)并純化與鑒定;采用戊二醛交聯(lián)法構(gòu)建磁靶向凝血蛋白(MNPs-tTF),體外分析其磁控靶向性及凝血效力。結(jié)果:成功構(gòu)建了磁靶向凝血蛋白MNPs-tTF,體外實(shí)驗(yàn)證實(shí)其保留有磁流體MNPs的磁控靶向性,以及凝血蛋白tTF的凝血效力。結(jié)論:成功構(gòu)建了磁靶向凝血蛋白MNPs-tTF,為血栓模型動(dòng)物的建立提供一種新工具。
[Abstract]:Aim: to construct a new type of chemoembolization agent, magnetic target coagulation protein (MTAP), which can induce thrombus in animals, so as to provide a new tool for the establishment of animal thrombus model. Methods: MNPs were synthesized by chemical coprecipitation method and their magnetic properties were determined by means of superconducting quantum interference magnetic intensity needle (SQUID) after dialysis. The truncated tissue factor (truncated tissue factor) was expressed, purified and identified by genetic engineering, and the magnetic target coagulation protein (MNPs-tTFA) was constructed by glutaraldehyde crosslinking method, and its magnetic targeting and coagulation potency were analyzed in vitro. Results: the magnetically targeted coagulation protein MNPs-tTFs was successfully constructed. It was confirmed by experiments in vitro that the MNPs-tTFs retain the magnetically controlled targeting of MNPs and the coagulation potency of tTF. Conclusion: MNPs-tTFs were successfully constructed, which provides a new tool for the establishment of thrombotic model animals.
