磷酸鈣納米體系用于順鉑前藥的運載

【摘要】:納米載藥體系能促進鉑類廣譜抗癌藥物的運輸和增強藥效,并保護作為前藥的四價鉑在到達細胞前不被還原.以磷酸鈣納米顆粒為基礎建立了一種生物兼容性高、制備方法簡單的四價鉑前藥運載體系.通過動態(tài)光散射、掃描電鏡和X射線衍射對顆粒粒徑和形貌進行了表征,并研究其細胞攝取效率和細胞毒性.實驗結果表明,該納米載藥體系的粒徑分布較窄、分散性較好、細胞攝取效率較高,并具有還原響應性,可在胞內還原劑的作用下釋放出順鉑,顯著提高了四價鉑前藥對腫瘤細胞的抑制作用.
[Abstract]:Nanopharmaceutical delivery system can promote the transport of platinum broad-spectrum anticancer drugs and enhance its efficacy, and protect tetravalent platinum as prodrug from reduction before reaching cells. Based on calcium phosphate nanoparticles, a tetravalent platinum prodrug delivery system with high biocompatibility and simple preparation method was established. The particle size and morphology were characterized by dynamic light scattering (DLS), scanning electron microscopy (SEM) and X-ray diffraction (XRD). The results showed that the nanoparticles had narrow particle size distribution, good dispersibility, high cell uptake efficiency and reduction response, and could release cisplatin under the action of intracellular reductant. The inhibitory effect of tetravalent platinum prodrug on tumor cells was significantly increased.
