【摘要】：目的:建立紫外分光光度法測(cè)定青霉素酶活力,并研究中國(guó)藥典青霉素酶活力單位與國(guó)際單位之間的關(guān)系。方法:利用青霉素與青霉噻唑酸在232 nm處的吸光系數(shù)不同,分別在30℃和37℃下,于232 nm處用紫外分光光度計(jì)監(jiān)控青霉素酶與一定量青霉素的反應(yīng)過(guò)程,通過(guò)反應(yīng)時(shí)間計(jì)算青霉素酶的活力;同時(shí)用中國(guó)藥典的碘量法進(jìn)行測(cè)定。結(jié)果:本文所用的紫外分光光度法測(cè)定結(jié)果與中國(guó)藥典規(guī)定的碘量法相比無(wú)顯著性差異(P=0.3020.05)。中國(guó)藥典青霉素酶活力單位(U)與國(guó)際單位(IU)的換算關(guān)系為:1 U=4.4×10~7 IU+2.6×10~5。結(jié)論:紫外分光光度法測(cè)定青霉素酶活力方法重復(fù)性好,操作簡(jiǎn)單,快速。建立的中國(guó)藥典青霉素酶活力單位與國(guó)際單位之間的關(guān)系使兩者具有可比性。
