【摘要】：目的:以斑馬魚為模型,評價(jià)并比較酒石酸長春瑞濱注射用乳劑與市售長春瑞濱水針劑(諾維本)的骨髓毒性、腸道毒性的變化情況。方法:以不同濃度酒石酸長春瑞濱注射用乳劑與水針劑靜脈注射給藥,觀察對斑馬魚紅細(xì)胞和中性粒細(xì)胞數(shù)量的影響以及對腸蠕動的影響,比較2種劑型的毒性情況。結(jié)果:長春瑞濱乳劑和水針劑均抑制紅細(xì)胞和中性粒細(xì)胞的形成,并呈現(xiàn)明顯的劑量效應(yīng);相同劑量的乳劑對中性粒細(xì)胞形成的抑制作用顯著低于水針劑(P0.001,P0.01);2種制劑對紅細(xì)胞形成的抑制作用無統(tǒng)計(jì)學(xué)差異;與5%葡萄糖注射液組相比,長春瑞濱水針劑顯著抑制腸蠕動(P0.01),并呈明顯的劑量效應(yīng);相同劑量乳劑組對腸蠕動僅產(chǎn)生輕微的抑制作用(抑制率:4%~7%),與5%葡萄糖注射液組相比無明顯差異。結(jié)論:與酒石酸長春瑞濱注射用水針劑相比,乳劑的骨髓抑制毒性和腸道毒性較低,提示乳劑具有降低長春瑞濱臨床不良反應(yīng)的優(yōu)勢。
[Abstract]:Objective: to evaluate and compare the changes of bone marrow toxicity and intestinal toxicity between vinorelbine tartrate emulsion and vinorelbine injection (Novibene). Methods: the effects of different concentrations of vinorelbine tartrate emulsion and water injection on the number of erythrocytes and neutrophils in zebrafish and the intestinal peristalsis were observed and the toxicity of the two formulations was compared. Results: both vinorelbine emulsion and water injection inhibited the formation of erythrocytes and neutrophils in a dose-dependent manner. The inhibitory effect of the same dose of emulsion on neutrophil formation was significantly lower than that of water injection P0.001, P0.01P0.01P0. 01, and compared with the 5% glucose injection group. Vinorelbine water injection significantly inhibited intestinal peristalsis (P0.01), and the same dose emulsion group had only slight inhibition (inhibition rate: 4% and 7%), and there was no significant difference compared with 5% glucose injection group. Conclusion: compared with vinorelbine tartrate injection, the bone marrow inhibition toxicity and intestinal toxicity of the emulsion are lower, suggesting that the emulsion has the advantage of reducing the clinical adverse reaction of vinorelbine.
