納米羥基磷灰石的制備及對(duì)阿霉素載藥性能的考察

【摘要】:目的合成藥物載體納米羥基磷灰石(nano-hydroxyapatite,nHAP),考察其對(duì)于模型藥物阿霉素的載藥性能。方法采用化學(xué)共沉淀法,以Ca(NO3)2·4H2O和(NH4)2HPO4分別為鈣、磷源,用氨水調(diào)控溶液p H值合成nHAP,考察溫度、反應(yīng)時(shí)間和陳化時(shí)間對(duì)于nHAP合成的影響。采用X射線衍射儀、掃描電鏡、紅外光譜儀、動(dòng)態(tài)光散射粒度測(cè)定儀對(duì)于產(chǎn)品的晶相、晶貌、化學(xué)組成和粒徑進(jìn)行表征。采用溶液吸附法考察nHAP用量、溶液質(zhì)量濃度和p H值對(duì)于nHAP負(fù)載阿霉素的影響。結(jié)果當(dāng)反應(yīng)溫度60℃、反應(yīng)時(shí)間2 h、陳化時(shí)間16 h時(shí),所制備的nHAP為長(zhǎng)(138±35)nm,寬(39±8)nm棒狀納米粒,晶體形貌規(guī)則,結(jié)晶良好。負(fù)載阿霉素的研究表明,所合成的nHAP載藥性能良好,載藥量達(dá)到42.43 mg·g~(-1)。結(jié)論 nHAP對(duì)于阿霉素具有良好的承載能力,可作為阿霉素的優(yōu)良載體。
[Abstract]:Aim to synthesize hydroxyapatite nano-hydroxyapatite nano-hydroxyapatite (HAPP) as drug carrier and to investigate its drug loading properties for adriamycin (adriamycin). Methods using Ca(NO3)2 4H2O and NH _ 4H _ 4HPO _ 4 as calcium and phosphorus sources, NH _ 4 and NH _ 4H _ 2HPO _ 4 were used to control pH value of solution to synthesize nHAPs by chemical coprecipitation method. The effects of temperature, reaction time and aging time on the synthesis of nHAP were investigated. The crystal phase, crystal appearance, chemical composition and particle size of the product were characterized by X-ray diffractometer, scanning electron microscope, infrared spectrometer and dynamic light scattering particle size analyzer. The effects of nHAP dosage, solution concentration and pH value on nHAP loaded adriamycin were investigated by solution adsorption method. Results when the reaction temperature was 60 鈩,
