細(xì)胞自噬對(duì)高三尖杉酯堿作用K562細(xì)胞的影響

【摘要】:目的:探討高三尖杉酯堿(HHT)單用或與自噬抑制劑3-甲基腺嘌呤(3-MA)聯(lián)用對(duì)K562細(xì)胞凋亡及自噬的影響。方法:K562細(xì)胞在HHT(10 ng/ml)作用下1-8 d,采用流式細(xì)胞術(shù)檢測細(xì)胞凋亡;采用RT-PCR法、Western blot法和透射電子顯微鏡檢測細(xì)胞自噬。結(jié)果:在HHT作用前期K562細(xì)胞凋亡率逐漸升高,而在后期則下降。HHT作用后,K562細(xì)胞的Beclin1表達(dá)及LC3Ⅱ/Ⅰ蛋白比值先下降后升高。HHT單用時(shí),激活型caspase-3蛋白表達(dá)前期逐漸上升,而在后期下降。與自噬抑制劑3-MA聯(lián)用組的K562細(xì)胞的Beclin1表達(dá)和LC3Ⅱ/Ⅰ的比值持續(xù)下降,而激活型caspase-3蛋白表達(dá)則逐漸升高。結(jié)論:HHT可誘導(dǎo)K562細(xì)胞凋亡,但HHT持續(xù)作用可誘導(dǎo)K562細(xì)胞發(fā)生自噬,與3-MA聯(lián)用則增強(qiáng)HHT的細(xì)胞毒作用。
[Abstract]:Aim: to investigate the effects of homoharringtonine (HHT) alone or in combination with autophagy inhibitor 3-methyladenine 3-MAon on apoptosis and autophagy in K562 cells.
