【摘要】：目的合成《歐洲藥典》中高血壓治療藥苯磺酸氨氯地平的雜質A、B、F、G、H,并對其進行了結構確證。方法以鄰苯二甲酸酐和乙醇胺為主要起始原料,經(jīng)酰胺化、O-烷基化、Knoevenagel縮合、Hantzsch環(huán)合、脫保護基等,得到所需雜質。結果合成了5個苯磺酸氨氯地平雜質,并通過M S、1H-NM R進行結構鑒定。結論制備了5個苯磺酸氨氯地平的雜質并對其來源進行了分析。該合成方法反應條件溫和、反應原料易得、操作簡單,產(chǎn)品具有高純度。
[Abstract]:Objective to synthesize the impurity of amlodipine benzenesulfonate in European Pharmacopoeia and its structure. Methods using phthalic anhydride and ethanolamine as the main starting materials, the necessary impurities were obtained by Knoevenagel condensation of Hantzsch cyclization and deprotection. Results five amlodipine benzenesulfonate impurities were synthesized and identified by MS 1H-NR. Conclusion five impurities of amlodipine benzenesulfonate were prepared and their sources were analyzed. The reaction conditions are mild, the raw material is easy to be obtained, the operation is simple, and the product has high purity.
