次黃嘌呤對(duì)小鼠脂質(zhì)代謝的影響

【摘要】:采用高脂飲食建立營(yíng)養(yǎng)性肥胖小鼠模型,連續(xù)口服給藥4周后收集血液和肝臟。通過(guò)熒光定量PCR技術(shù)對(duì)小鼠肝臟中AP-2、PPAR-γ和LPL基因的mRNA相對(duì)表達(dá)量進(jìn)行分析。以小鼠體質(zhì)量增長(zhǎng)率、脂肪質(zhì)量、總膽固醇、甘油三脂等和小鼠肝臟中AP-2、PPAR-γ和LPL基因的mRNA的相對(duì)表達(dá)量為指標(biāo),觀察次黃嘌呤的減肥作用。結(jié)果表明:經(jīng)給藥后,給藥組對(duì)小鼠體內(nèi)脂肪有不同程度的干擾作用,小鼠脂肪質(zhì)量顯著減少(P0.05),肥胖小鼠血清總膽固醇和甘油三酯水平降低,LDL-C和LPS的水平也得到明顯改善(P0.05)。其中次黃嘌呤低劑量給藥組和次黃嘌呤中劑量給藥組對(duì)于降低肥胖小鼠的脂肪質(zhì)量均有效果。
[Abstract]:Nutritional obesity mice model was established by high fat diet. Blood and liver were collected after oral administration for 4 weeks. The relative expression of AP-2PPAR- 緯 and LPL genes in mouse liver was analyzed by fluorescence quantitative PCR. The relative expression of AP-2PPAR- 緯 and LPL gene in the liver of mice and the relative expression of AP-2PPAR- 緯 and LPL gene were used as indexes to observe the weight loss effect of Hypoxanthine on the growth rate of body mass, fat mass, total cholesterol, triglyceride and so on. The results showed that: after administration, the body fat of mice was interfered with in different degree in the administration group, the fat quality of mice significantly decreased P0.05N, the serum total cholesterol and triglyceride levels of obese mice decreased the levels of LDL-C and LPS, and the levels of P0.05G were also significantly improved. The low dose of Hypoxanthine and the medium dose of Hypoxanthine were effective in reducing the fat quality of obese mice.
