【摘要】：目的探討埃羅替尼對(duì)小鼠角膜上皮組織引起的組織病理學(xué)及超微結(jié)構(gòu)改變的影響。方法30只雄性6~8周齡BALB/c小鼠,隨機(jī)分為對(duì)照組(12只)和實(shí)驗(yàn)組(12只),6只不作處理為空白對(duì)照。實(shí)驗(yàn)組使用20μmol·L~(-1)埃羅替尼液滴眼,對(duì)照組采用PBS滴眼,每天4次。分別在干預(yù)后1 d、7 d、14 d對(duì)各組小鼠進(jìn)行熒光素鈉(fluorescent,FL)染色及評(píng)分。干預(yù)后14 d取小鼠眼球,光學(xué)顯微鏡及電子顯微鏡下觀察角膜上皮及細(xì)胞的結(jié)構(gòu)變化。提取角膜蛋白進(jìn)行Western Blot檢測。結(jié)果干預(yù)前,實(shí)驗(yàn)組和對(duì)照組FL評(píng)分比較差異無統(tǒng)計(jì)學(xué)意義(P0.05)。在干預(yù)后1 d、7 d、14 d,實(shí)驗(yàn)組FL評(píng)分較干預(yù)前明顯升高,差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05),而對(duì)照組FL評(píng)分與干預(yù)前無明顯改變,差異均無統(tǒng)計(jì)學(xué)意義(均為P0.05);兩組間FL評(píng)分在干預(yù)后1 d相比,差異無統(tǒng)計(jì)學(xué)意義(P0.05);兩組間FL評(píng)分在干預(yù)后7 d、14 d相比,差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05)。實(shí)驗(yàn)組小鼠角膜上皮細(xì)胞排列紊亂,層數(shù)增加;電鏡下見角膜上皮表層細(xì)胞形態(tài)不規(guī)則、脫落,微絨毛減少、消失;表層上皮細(xì)胞的橋粒、半橋粒連接數(shù)目明顯減少。實(shí)驗(yàn)組角膜表皮生長因子受體表達(dá)明顯發(fā)生變化,兩組表皮生長因子受體比較差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論埃羅替尼會(huì)引起小鼠角膜上皮的組織結(jié)構(gòu)及細(xì)胞超微結(jié)構(gòu)損壞。其機(jī)制可能是通過抑制表皮生長因子受體的活化來影響角膜上皮的。
[Abstract]:Objective to investigate the effects of erotinib on histopathological and ultrastructural changes induced by corneal epithelium in mice. Methods Thirty male BALB/c mice aged 6 to 8 weeks were randomly divided into control group (n = 12) and experimental group (n = 12). The experimental group was treated with 20 渭 mol L ~ (-1) Erotini drops, while the control group was treated with PBS drops 4 times a day. The mice in each group were stained with sodium fluorescein fluorescein FLL on the 7th day and 14th day after intervention. The cornea epithelium and cell structure were observed by optical microscope and electron microscope 14 days after intervention. Cornea protein was extracted for Western Blot detection. Results before intervention, there was no significant difference in FL score between the experimental group and the control group (P 0.05). The scores of FL in the experimental group were significantly higher than those before the intervention on the 1st day, 7th day and 14th day after the intervention (all P 0.05), but the FL score of the control group had no significant change compared with that before the intervention. There was no significant difference in FL score between the two groups (P 0.05), and there was no significant difference in FL score between the two groups on the 1st day after intervention (P 0.05), and there was significant difference in FL score between the two groups at 7 d and 14 d after intervention (all P 0.05). In the experimental group, the corneal epithelial cells arranged disorderly and the number of layers increased; under the electron microscope, the corneal epithelial surface cells were irregular in shape, shedding off, the microvilli decreased and disappeared; the desmosome and semi-desmosome junctions of the surface epithelial cells decreased significantly. The expression of epidermal growth factor receptor (EGF) in the experimental group was obviously changed, and the difference between the two groups was statistically significant (P 0.05). Conclusion Erotini can damage the tissue structure and cell ultrastructure of corneal epithelium in mice. The mechanism may affect corneal epithelium by inhibiting the activation of epidermal growth factor receptor (EGF).
